摘要
以铁皮石斛原球茎为试材,应用3L气升式生物反应器,研究了反应器不同培养时期、通气方式和接种量对铁皮石斛原球茎生物量及有效物质积累的影响,并比较了铁皮石斛不同来源材料的多糖与生物碱含量。结果表明:铁皮石斛原球茎的生物量及多糖和生物碱的积累量均在培养至30d时达到最大值,在反应器培养过程中逐步调节通气量由0.1vvm至0.3vvm,可提高原球茎的生物量及有效物质的含量,接种量为20-25g/L均可获得较高的原球茎生物量且有效物质含量和产量也较高,铁皮石斛原球茎的多糖和生物碱含量均高于1年生植株和组培苗。因此,铁皮石斛原球茎反应器培养30d,培养期间将通气量由0.1vvm调节至0.3vvm,接种量为20-25g/L有利于大量生产原球茎及其有效物质。
Taking D.candidumPLB as experiment material,the effect of culture days,aeration method and inoculation density on accumulation of biomass and bioactive compounds of D.candidumPLB were studied,by using 3 Lair-lift bioreactor.Additionally,the content of polysaccharide and alkaloid from different sources were compared.The results showed that the accumulation of biomass and bioactive compounds achieved the highest on the 30d,when the air volume was adjusted from 0.1vvm to 0.3vvm during bioreactor culture and inoculation density was 20-25g/L.The content of polysaccharide and alkaloid in D.candidumPLB was higher than cultivation plants and tissue culture seedlings.It was conducive to accumulation of biomass and bioactive compounds of D.candidumPLB in that conditions.
