摘要
目的:探讨miR-939在人肺癌细胞株A549中的表达及增强miR-939表达对肺癌细胞周期的影响。方法:提取15对肺癌、癌旁正常组织标本的RNA,用qPCR法检测miR-939的表达。将miR-939前体转染入A549中,实验分为三组,即前体转染组(P组)、转染试剂阴性对照组(N组)、转染试剂空白对照组(C组),分别以qPCR法检测miR-939前体转染后三组细胞内miR-939的表达水平。用MTT法测定细胞生长曲线。PI染色法检测转染后三组细胞周期变化情况。结果:相对肺癌癌旁正常组织,86.7%肺癌组织中miR-939呈低表达(P<0.01)。P组中miR-939表达水平较N组、C组都显著升高(P<0.01)。转染48h后,P组较N组、C组,G1期细胞百分比明显增多,S期细胞百分比明显减少(P<0.05)。结论:miR-939在肺癌组织中呈低表达,上调A549细胞中miR-939的表达,能够诱导细胞周期阻滞。提示miR-939在肺癌的发生发展过程中有可能起着重要作用。
Objective:To investigate the expression of miR -939 in human lung cancer cell line A549, and to ex- plore its influence on the cell cycle when the expression of miR -939 is enhanced. Methods: RNAs of 15 pairs of specimens of lung cancer and adjacent non - tumorous tissues were extracted, and then the expressions of miR - 939 were detected by quantitive real - time PCR. miR - 939 precursors were transfected into the cell line A549, the cells were divided into three groups :Precursors transfected group ( group P), the negative control group ( group N ) and the bank control group ( group C). Then the expressions of miR - 939 were detected by qPCR, the changes of the cell cy- cle were determined by PI incorporation. Results : Compared with adjacent non - tumorous tissues, the expression of miR -939 was lower (P 〈 0.0! ) in 86.7% of lung cancer. Compared with the group N and the group C, the expres- sion of miR - 939 in the group P was increased markedly. After d8 hours transfection, to group P, the percentage of cells in G1 phase was increased. The percentage of cells in S phase was decreased. Conclusion:The expression level of miR - 939 is down - regulated in lung cancer. With up - regulation of miR - 939 expression in the cell line A549, cell cycle can be arrested. It may correlate with the formation and progression of lung cancer.
