锌转运蛋白4cDNA稳定转染对前列腺癌DU145细胞株生物学行为的影响

摘要 目的探讨锌转运蛋白(ZIP)4对前列腺癌细胞系生物学行为的影响。方法比较3种不同的前列腺癌细胞系(22RV1、DU145、PC3),选择ZIP4低表达的细胞株,转染构建ZIP4过表达的前列腺癌细胞系,进一步应用MTT、Transwell及细胞划痕等试验检测ZIP4过表达后对前列腺癌细胞系增殖活性及侵袭转移能力的影响。结果建立稳定表达ZIP4的DU145前列腺癌细胞系;转染后的DU145-ZIP4细胞系的增殖活性及侵袭能力与转染空载体组相比均显著下降(P<0.05),但其迁移能力与转染空载体组相比未见明显变化(P>0.05)。结论 ZIP4基因对前列腺癌细胞系的增殖活性及侵袭能力有显著抑制作用,ZIP4可能是前列腺癌的一个抑癌基因。 Objective To investigate the effects of zinc transporter (ZIP) 4 on the biological behavior of prostate cancer cell lines.Methods The expression of ZIP4 in three different prostate cancer cell lines (22RV 1,DU 145,PC3) was detected.DU 145,which shows the lowest expression of ZIP4,was selected for next studies.Stable transfection of ZIP was consaucted in the DU145 cell line,and then different methods including MTT,Transwell,and wound healing were applied to detect the biological behavior of DU 145-ZIP4 cell line.MTT was used to detect the proliferation ; transwell was used to detect invasion and metastasis ability ; wound healing was used to investigate the migration.Results Stable prostate cancer cell line expressing ZIP4 was established.The proliferation and invasion ability of DU 14-ZIP4 cell line were significantly decreased compared with that of control group (DU 145 cell and DU 145 transfected with empty vector) (P < 0.05) ; but the migration ability showed no significant change between DU 145-ZIP4 and the control groups (P > 0.05).Conclusion Overexpression of ZIP4 has significant inhibitory effect on the proliferation and invasive ability in prostate cancer cell line DU 145-ZIP4,which suggested that ZIP4 may serve as a tumor suppressor gene in prostate cancer.
