摘要
目的观察多囊卵巢综合征(polycystic ovary syndrome,PCOS)患者外周血中Th17细胞与FOXP3+CD25highCD4+调节性T(Treg)细胞比例及相关因子表达的变化并探讨其意义。方法取8例PCOS患者及12例健康妇女外周血,流式细胞术检测Th17和Treg细胞占CD4+T细胞的比例;实时定量PCR测定外周血单个核细胞(PBMC)中Th17和Treg细胞相关因子IL-17、RORγt、FOXP3、TGF-β、IL-1β及IL-6 mRNA的相对表达量。结果 PCOS患者外周血中Th17细胞的比例明显高于健康妇女(P<0.01),Treg细胞的比例较健康妇女有增高趋势但差异无统计学意义(P>0.05)。PCOS患者PBMCs中,IL-17、RORγt、FOXP3、TGF-β的mRNA相对表达量明显高于健康妇女(P<0.01),而IL-1β则明显低于健康妇女(P<0.01),IL-6差异无统计学意义(P>0.05)。结论 PCOS患者可能处于一种Th17细胞明显增多的炎性状态,这可能参与了PCOS的发生发展,但其中细胞因子的作用尚待进一步研究。
Objective To observe the changes of Th17 and FOXP3^ + CD25^highCD4 ^+ regulatory T (Treg) cells and Thl7/Treg-related factors in peripheral blood of patients with polycystic ovary syndrome (PCOS), and explore the effects of peripheral Th17 and Treg cells on pathogenesis of PCOS. Methods The blood samples were collected from 8 PCOS patients and 12 healthy women. The ratios of Thl7 and Treg cells in the peripheral blood samples were examined with flow cytometry. The relative mRNA quantities of IL-17, RORγt, FOXP3, TGF-β, IL-1β and IL-6 were examined by real-time PCR. Results Compared with healthy controls, the level of Th17 cells in PCOS patients was much higher ( P 〈 0.01 ), while that of Treg cells was similar ( P 〉 0.05 ). The relative mRNA quantities of IL-17, RORγt, FOXP3 and TGF-β in PCOS patients were much higher than those in healthy controls (P 〈 0.01 ), while the relative mRNA quantity of IL-6 was similar with healthy controls (P 〉 0.05 ), amt IL-1β was significantly lower than healthy controls ( P 〈 0.01 ). Conclusion In PCOS patients Th17-dominant inflammatory status may involve in the pathogenesis of disease.
