缬沙坦对球囊损伤后大鼠主动脉组织TLR4表达影响 被引量:3

THE EFFECTS OF VALSARTAN ON EXPRESSION OF TOLL-LIKE RECEPTOR 4 IN RAT AORTIC TISSUE AFTER BALLOON INJURY

摘要 目的通过观察大鼠主动脉内皮球囊损伤后血管内膜增生及Toll样受体4(TLR4)表达的变化,探讨TLR4在再狭窄中的作用及缬沙坦对其干预的影响。方法建立球囊损伤大鼠主动脉内皮模型,并将大鼠随机分为对照组、手术组和缬沙坦治疗组,每组分别于术后14及28d取主动脉组织,苏木精-伊红染色观察血管内皮厚度的变化,逆转录聚合酶链反应检测主动脉组织TLR4mRNA的表达。结果术后14及28d手术组大鼠主动脉血管内膜较对照组均显著增厚(F=114.46、118.56,q=20.65、21.05,P<0.05),TLR4mRNA表达较对照组均显著升高(F=281.97、245.02,q=31.88、31.30,P<0.05)。术后14及28d缬沙坦治疗组内膜增生程度均轻于手术组(q=5.49、5.68,P<0.05),TLR4mRNA表达较手术组均显著降低(q=6.81、16.35,P<0.05)。结论缬沙坦可抑制大鼠主动脉内皮损伤后的血管内皮增生,可能与其抑制TLR4的表达而减轻炎症反应有关。 Objective To investigate the intimal hyperplasia and expression of toll-like receptor 4(TLR4)after aortic balloon injury in rats,and explore the effects of TLR4on restenosis and the action of valsartan on it. Methods A model in rats with aortic balloon injury was created.The rats were randomized to control group,operation group and valsartan group.The aortic tissues were taken from rats in each group on days 14and 28after surgery.The thickness of vascular endothelium was observed with HE stain,the expression of TLR4mRNA was detected by using RT-PCR. Results Compared with the control group,the thickness of vascular intima in the operation group was much obvious(F=114.46,118.56;q=20.65,21.05;P〈0.05),and the expression of TLR4mRNA increased(F=281.97,245.02;q=31.88,31.30;P〈0.05).Compared with the operation group,the intimal hyperplasia in the valsartan group was slighter(q=5.49,5.68;P〈0.05),and the expression of TLR4mRNA was lower(q=6.81,16.35;P〈0.05). Conclusion Valsartan can inhibit vascular endotheliosis in rats caused by aortic endothelial damage,which is likely to be associated with inhibition of the expression of TLR4and thus reduce the inflammatory reaction.
