摘要
目的 观察粉防己碱(Tet)对β-葡聚糖(β-glucan)诱导的巨噬细胞增殖、胞内游离钙及环磷酸腺苷(cAMP)表达的影响.方法 以终浓度为100 μg/ml的β-glucan刺激RAW264.7细胞,建立细胞模型.MTT法检测不同浓度Tet(0,0.01,0.1,1,10,100 μmol/L)对RAW264.7细胞增殖的影响;动态检测Tet(0.01,0.1,1μmol/L)剂量组对胞内游离钙和cAMP表达的影响.结果 与对照组比较,Tet(0.01,0.1,1μmol/L)剂量组A值升高(P<0.01);胞内游离钙在Tet作用5 min时表达增加(t=4.18,4.17,3.40,P均<0.01);cAMP浓度在1 min时明显升高(t=3.33,3.84,3.18,P均<0.01).Tet>10 μmol/L时,A值下降(P<0.01).结论 适当浓度的Tet可以促进β-glucan诱导的RAW264.7细胞生长,协同βglucan调节细胞内游离钙及cAMP的表达.
Objective To observe the effect of tetrandrine (Tet) on β glucan induced macrophage proliferation,the expression of [Ca2+]i and cAMP.Methods With the 100 μg/ml β glucan stimulates RAW264.7 cell model.MTT to detect of Tet (0,0.01,0.1,1,10 100 μmol/L) on the proliferation of RAW264.7 cell; dynamic detection of Tet (0.01,0.1,1 μmol/L) on [Ca2+]i and cAMP concentration.Results Compared with the control group,Tet (0.01,0.1,1 μmol/L) group A values in creased (P<0.01) ;intracellular [Ca2+]i expression increased in 5 min (t=4.18,P<0.01 ; t=4.17,P<0.01 ; t=3.40,P< 0.01) ; cAMP concentration increased significantly at 1 min (t=3.33,P<0.01 ; t=3.84,P<0.01 ; t=3.18,P<0.01).A values decreased significantly when Tet>10 μmol/L (P<0.01).Conclusion Suitable concentration of Tet can promote the growth of β-glucan induced RAW264.7 cells,and collaborative β glucan regulates the expression of [Ca2+] and cAMP.
