高原植物芜菁挥发油成分降血糖作用研究

Study on Hypoglycemic Action of Plateau Brassica rapa Volatile Oil Components

目的:探讨高原植物芜菁挥发油成分对糖尿病小鼠动物模型的降血糖作用。方法:以超临界二氧化碳萃取仪提取芜菁挥发油成分。150只雄性小鼠采用高脂高糖喂养4周后,辅以连续3 d ip链尿佐菌素(STZ,100 mg·kg-1)造小鼠糖尿病模型,采用血糖测定仪尾静脉取血测空腹血糖,以血糖>11.0 mol·L-1为造模成功小鼠。将入选的40只小鼠随机分为正常对照组,模型组,芜菁挥发油高、低剂量(40,20 g·kg-1)组,阳性组(盐酸二甲双胍0.2 g·kg-1),共5组,ig每日1次,连续14 d,实验期间分别于造模前、造模后、用药后3,7,14 d剪尾取血检测血糖。实验结束后,小鼠摘眼球取血,取全血测定糖化血红蛋白,分离血清测定血清胰岛素水平,剖取新鲜的胸腺、脾脏,计算脏器指数,观察各组大鼠的血糖、脏器指数、糖化血红蛋白及胰岛素水平的变化。结果:芜菁挥发油成分可显著降低2型糖尿病模型小鼠的血糖(P<0.01),降低小鼠糖化血红蛋白(P<0.01,P<0.05),升高血清胰岛素水平(P<0.01)。结论:芜菁挥发油成分对STZ造成小鼠糖尿病模型有较明显的降血糖作用。

Objective：To discuss the hypoglycemic action of plateau plant turnip volatile oil constituents.Method：The supercritical carbon dioxide extraction was used to extract turnip volatile oil components.The animal model of diabetic mice was induced by feeding high serum glucose and serum lipid fro 4 weeks and injection of streptozotocin （100 mg ·kg）.The blood sugar level was detected.Forty mice whose blood glucose level was between 15-17 mmol ·L-1 were randomly divided into model group,turnip volatile oil （high,low dose group）,positive control group （metformin hydrochloride）,and normal control group,altogeher 5 groups,the intragastric administration was conducted once per day for 14 days.During the experimental period,the blood glucose level was measured after 3,7,14 days of administration.After the experiment,blood samples were collected for measurements of glycosylated hemoglobin and serum insulin level.The fresh spleen and thymus gland were collected for computing organ index.Result：Turnip volatile oil components could significantly reduce blood glucose and glycosylated hemoglobin,and increase serum insulin levels.Conclusion：Turnip volatile oil components significantly reduce blood glucose.
