铁皮石斛对运动疲劳小鼠能量代谢和CNTFmRNA表达的影响 被引量:9

Effects of Dendrobium officinale on Energy Metabolism and Expression of CNTF mRNA in Athletic Fatigue Mice

摘要 目的:观察铁皮石斛对运动疲劳小鼠能量代谢和睫状神经营养因子(ciliary neurotrophic factor,CNTF)mRNA表达的影响。方法:清洁级雄性昆明种小鼠60只随机分为正常对照组(简称对照组)、铁皮石斛低、中、高剂量组共4组。每组15只。对铁皮石斛低、中、高剂量组按0.75,1.5,4.5 g·kg-1分别给予铁皮石斛灌胃,对照组灌胃相同体积的生理盐水。每天1次,连续30 d。然后通过负重游泳实验,观察小鼠负重游泳时间,检测血清尿素、肌糖原、肝糖原及乳酸含量和CNTF mRNA表达。结果:与对照组负重游泳时间比较,铁皮石斛低、中、高剂量组均显著延长(P<0.05或P<0.01)。与对照组血清尿素、乳酸含量比较,铁皮石斛低、中、高剂量组均显著降低(P<0.05或P<0.01)。与对照组肝糖原、肌糖原含量比较,铁皮石斛中、高剂量组均显著升高(P<0.05或P<0.01)。与对照组CNTF mRNA表达量比较,铁皮石斛低、中、高剂量组均显著升高(P<0.05或P<0.01)。结论:铁皮石斛通过提高机体糖原储备、减少血清尿素含量、减轻乳酸堆积,同时提高CNTF mRNA表达,从调节能量代谢和营养肌肉两方面发挥功效可能是其抗疲劳的机制之一。 Objective:To investigate the effects of Dendrobium officinale on energy metabolism and expression of ciliary neurotrophic factor (CNTF) mRNA in athletic fatigue mice.Method:Sixty male Kunming mice were divided randomly into control group,D.officinale low-dose group,middle-dose group,high-dose group.The low-dose group,middle-dose group and high-dose group were intragastricaly (ig) administrated with D.officinale respectively according to 0.75,1.5,4.5 g · kg-1 while control group was ig given same volume normal saline,once a day for 30 days.The weight carrying swimming test were used to observe exhaustive weight carrying swimming time and to examine levels of serum urea,muscle glycogen,hepatic glycogen,lactic acid and the expression of CNTF mRNA.Result:Compared to control group,exhaustive weight carrying swimming time in low-dose group increased (P < 0.05),and in both middle-dose group and high-dose group increased significantly (P<0.01).Compared to control group,the levels of serum urea and lactic acid in both low-dose group and middle-dose group decreased (P < 0.05),and in high-dose group decreased significantly (P < 0.01).Compared to control group,the levels of muscle glycogen and hepatic glycogen in middle-dose group increased (P < 0.05)and in high-dose group increased significantly (P < 0.01).Compared to control group,the expression of CNTF mRNA in low-dose group increased (P < 0.05),and in middle-dose group and high-dose group increased significantly (P <0.01).Conclusion:D.officinale could increase glycogen store,decrease the level of serum urea and lactic acid accumulation,meantime,could up-regulate the expression of CNTF mRNA,which may be one of mechanisms that D.officinale alleviate athletic fatigue by regulating energy metabolism and nourishing muscle.
