摘要
目的观察大蒜素对肾细胞癌ketr-3细胞生长、细胞凋亡及细胞周期的影响。方法不同浓度的大蒜素作用于体外培养的ketr-3细胞。倒置显微镜下观察ketr-3细胞的形态学变化,采用四唑盐比色法(MTT)及流式细胞术(FCM)检测不同时期ketr-3细胞形态变化、增殖以及细胞凋亡的作用。结果大蒜素对肾细胞癌ketr-3细胞生长具有抑制作用,在作用24、48、72h点上不同浓度的大蒜素对ketr-3细胞的抑制率之间差异有统计学意义(P<0.05)。大蒜素作用24 h即可检测到凋亡细胞,0.05 mg/m L、0.1 mg/m L、0.3mg/m L、0.5mg/m L药物组凋亡率分别为9.21%、27.43%、37.72%、46.52%,1mg/m L组的凋亡率最高,达到62.21%,而对照组仅为3.13%,差异有统计学意义(P<0.05)。结论大蒜素能够诱导ketr-3细胞凋亡,阻滞细胞周期,抑制细胞增殖。
Objective To investigate the effect of allicin on growth, apoptosis and cell cycle of human renal cell carcinoma(ketr-3) cell. Methods Effect of different concentrations of allicin on the proliferation of ketr-3 cells. Morphological changes in ketr-3 ceils was observed under inverted microscope, The effects of allicin on morphology, proliferation and apoptosis of ketr-3 cell at different periods were detected using four MTT assay (MTT) and flow cytometry (FCM).Results Allicin could inhibit the growth of carcinoma kerr-3 cell showing significantly different inhibitory effect 24,48, 72h after action with allicin (P〈0.05). Apoptotic cells could be detected 24 hours after action with allicinat concentration of 0.05 mg/mL, 0.1 mg/mL, 0.3mg/mL, 0.Smg/mL with the apoptosis rates of 9.21% , 27.43% , 37.72% , 46.52%, The apoptosis in lmg/mL group was 62.21% ,being the highest, while that of the control group was only 3.13%. Conclusion Allicin possesses significant effects on the proliferation of ketr - 3 and inducing the apoptosis and arrestment of cell cycle.
