益气活血利水方对慢性心力衰竭大鼠心肌重构的影响

目的:观察益气活血利水方益心泰对慢性心力衰竭大鼠心肌重构的影响。方法:采用冠状动脉结扎法建立慢性心力衰竭(CHF)大鼠模型,将造模成功的Wistar雄性大鼠随机分为模空组、益心泰高剂量组、益心泰中剂量组、益心泰低剂量组和科索亚组,另设正常对照组。益心泰低、中、高剂量组分别予以益心泰灌胃,剂量分别为2.6g/(kg·d)、5.2 g/(kg·d)、10.4 g/(kg·d)。科索亚组灌胃剂量为5.21g/(kg·d)。模型组灌服同等体积的生理盐水。每天上午灌胃1次,共8周。正常对照组不做任何处理。8周后检测心衰大鼠心脏超声指标、心肌重构指标。同时采用H-E染色和Massion染色观察左心室心肌形态结构变化。结果:与模空组比较,益心泰低、中、高剂量组和科索亚组LVIDs、LVIDd均显著降低,LVEF、LVFS明显升高(P<0.05)。而益心泰高剂量组与科索亚组各项指标比较差异无统计学意义(P>0.05)。结论:益心泰可改善心衰大鼠的心肌重构,延缓心衰的进展。

Objective： To observe the effect of replenishing qi, activating blood and promoting urination decoction Yixintai on myocardial remodeling of chronic heart failure rats. Method： CHF rat models were established by coronary artery ligation. The successful modeling male Wistar rats were divided into 6 groups randomly, the blank model group, the low, middle and high dosage of Yixintai group and Losartan group. Moreover, a normal group was used for comparison. The low, middle and high dosage of Yixintai groups were taken Yixintai by intragastric administration with the dosage of 2.6g/（kg · d）, 5.2 g/（kg · d）, 10.4 g/（kg · d）. The dosage of Losartan group for intragastric administration was 5.21g/（kg · d）. The model group received the same volume of saline. The rats received intragastric administration once every morning, for the next 8 weeks. The normal group received no treatment. After 8 weeks, the echocardiography index and myocardial remodeling index of heart .failure rats were examined. Meanwhile, the changes of left ventricular myocardial morphology and structure were observed by HE staining and Massion staining method. Result： Compare with LVIDs and LVIDd of the model group, that of the low, middle and high dosage of Yixintai group and Losartan group all decreased significantly （ P〈0.05 ） . Compare with LVEF and LVFS of the blank model group, that of the low, middle and high dosage of Yixintai group and Losartan group all increased significantly （ P〈0.05 ） . There was no statistical significance in each index between the high dosage of Yixintai group and Losartan group （ P〉0.05 ） . Conclusion： Yixintai could improve myocardial remodeling of heart failure rats and delay the progress of heart failure.
