电化学发光法检测鲎素

A highly sensitive electrochemiluminescence method for the determination of tachyplesin

摘要 基于鲎素(TCP)能强烈抑制钌联吡彤N,N-二丁基乙醇胺(Ru(bpy)3 2+/DBAE)体系的电化学发光(ECL)信号,建立了检测鲎素的方法。在最优实验条件下,Ru(bpy)3 2+/DBAE体系的ECL淬灭值△I EcL与TCP质量浓度的对数log P呈良好的线性关系,线性范围为3.0×10 -10~1.5×10 -8g/mL,检出限达50pg/mL(S/N=3)。方法用于鲎试剂样品分析,鲎素的加标回收率为93.0%~107.6%. A highly sensitive method for the determination of tachyplesin( TCP)was developed based on its strong eleetrochemiluminescence (ECL) inhibition of Ru (bpy) 3 2 +/DBAE system. Under the optimized conditions, the quenched EcL intensity versus the logarithm of the concentration of tachyplesin was linear over a concentration range from 3.0 × 10-10 μg/mL to 1.5 × 10 -8 g/mL. The limit of detection (,LOD) was found to be 5.0 × 10-11 g/mL (S/N = 3 ). And this method was successfully applied to the estimation of tachyplesin in limulus reagent, with the recoveries in the range from 93.0% to 107.6%.
