承德产黄芩不同采收期茎、叶中野黄芩苷的含量测定

摘要 目的测定并比较河北省承德隆化县黄芩GAP种植基地所产的黄芩不同采收期茎、叶中的野黄芩苷的含量。方法采用HPLC,色谱柱为AgilentZorbaxSB-C18(4.6mm×250mm,5μm),流动相为甲醇-水-甲酸(53:47:0.1),流速为0.8ml·mn-1,检测波长为335nm。结果野黄芩苷在0.24~0.72μg内呈良好线性关系(r=0.9999),平均加样回收率99.69%.RSD0.93%(n=6)。经比较发现,此批采的承德产黄芩的叶中野黄芩苷含量在6-10月总体呈下降趋势,2012年6月20日的黄芩的叶中野黄芩苷含量最高,达到2.68%。黄芩的茎中野黄芩苷含量在6-10月总体呈较平稳趋势,2012年9月20日黄芩的茎中野黄芩苷含量最高,达到0.74%。结论该方法简便、稳定可靠,重现性好,不同采收期的黄芩中野黄芩苷的含量存在差异并有消长规律。 Objective To compare the content of scutellarin from which was produced at Hebei Provincial GAP planting area of Scutellaria baicalensis( Longhua country) at different collecting time. Methods Diosgenin was separated on a Zorbax SB -C18 (4.6 mm χ250 mm,5 ~m) column and detected at 335nm at the flow rate of 0.8 mL . min- ~ using a mixture of methanol - H20 - formic acid (53 : 47 : 0.1 ). Results The standard curve was linear in the range of 0.24 - 0.72 tLg with correlation coefficient 0.9999. The average recovery of baicalin was 99.69% with RSD O. 93% ( n =6 ). The total content of scutellarin from Scutel- laria baicalensis leaves of Chengde from June to October decreased continuously,and the maximum value was June 20,2012 ,up to 2.68%. The total content of scutellarin from Scutellaria baicalensis leaves of Chengde from June to October showed a trend of rel- atively stable as a whole, and the maximum value was September 20,2012, up to 0.74%. Conclusion The method was simple, sta- ble and reliable, reproducible,there were differences in the contents of baicalin and changed regularly.
