摘要
目的基于NF-κB通路探讨薯蓣皂苷含药血清对IL-1β、IL-17和TNF-α诱导大鼠滑膜细胞株RSC-364的影响。方法采用IL-1β、IL-17和TNF-α联合刺激RSC-364建立类风湿性关节炎细胞模型,制备薯蓣皂苷片和雷公藤多苷片含药血清,应用RT-PCR、Western-Blot检测各组中NF-κBP65 mRNA/蛋白、I-κB/蛋白的表达结果。结果与空白组比较,模型组NF-KBP65mRNA/-NR1表达增加,I-κB mRNA/-NR1减少,差异具统计学意义(P<0.05);与模型组比较,中药组、对照组NF-κBp65减少,I-κB mRNA增加,I-κB-NR1蛋白增加,I-κB-NR1蛋白表达明显减少。结论薯蓣皂苷片含药血清可能通过调控I-κB来抑制转录因子NF-κB p65的产生,进而激活NF-κB信号转导通路。
Objective Based on NF - KB pathway to investigate effect of serum contained dioscin on IL - 113,IL - 17 ~ TNF - ct induced rat synovial cell strain RSC -364. Methods Combined stimulated RSC -364 by IL- 1 [3,IL- 17 and TNF-c t, to estab- lished Rheumatoid arthritis cell models, prepared serum contained diosein tablets and tripterygium glycosides tablets, adopted RT -PCR,WESTERN- BLOT to test NF - KBP65 mRNA/ albumen's, I - KB/ albumen's expression of resuhsResults compared with blank group, model group~ NF - KBP65mRNA/- NR1 expression increased, I - KB mRNA/- NR1 reduced, the differences had statistical significations (p 〈 0.05 ) ;compared with model group, Chinese medicine group's, control group's NF - KBP65 re- duced, I - KB mRNA increased, I - KB - NR1 albumen increased, I - KB - NR1 albumen expression obviously reduced. Conclu- sion Serum contained dioscin maybe suppress transcription factor by adjust I - KB, and then activate NF - KB signal transduction pathway.
