摘要
目的观察桃红四物汤对寒凝血瘀模型大鼠HO/CO系统的影响,探讨桃红四物汤治疗寒凝血瘀型妇科疾病的作用机制。方法将大鼠置于0-1℃冰水中造成寒凝血瘀大鼠模型,采用血红蛋白结合及联二亚硫酸盐还原法测定血浆CO活性;采用免疫组织化学sP法检测卵巢组织HO-1、HO-2蛋白表达;采用原位杂交法检测卵巢组织HO-1、HO-2mRNA表达。结果桃红四物汤组大鼠血浆COHb活性较模型组增强(P〈0.05)。卵巢颗粒细胞、黄体细胞HO-1、HO-2蛋白表达,HO-1、HO-2mRNA表达加强(P〈0.01或P〈0.05)。结论桃红四物汤治疗寒凝血瘀型妇科疾病的作用与调节HO/CO系统有关。
Objective To observe the effect of modified TaoHongSiWu Decoction on model rat of blood stasis due to cold accumu- lation in the expression of HO / CO system and to explore its mechanism of treatment on Gynecological disease. Methods The rat model of blood stasis due to cold accumulation was induced by keeping them in water at 0 - 1 ~C. The CO activity was detected by the methods of protein binding and dithionite reduction; the protein expression of HO - 1, HO - 2 in ovary was determined by Im- munohistochemistry analysis ; in situ hybridization histoehemistry analysis was used to reveal the mRNA expression of HO - 1, HO - 2 in ovary. Results Compared with the model group, the activity of plasma COHb was significantly increased, while the protein expression and mRNA expression of HO - 1 and HO - 2 in ovarian granulosa cells and lnteal cells were enhanced, after adminis- tration with Modified TaoHongSiWu Decoction. Conclusion The therapeutical effect of TaoHongSiWu Decoction on Gynecological disease was related to regulate the expression of HO / CO system.
