Antidepressant mechanisms in mTOR signaling cascade

摘要 尸检研究发现,MDD患者前额叶皮层存在大量mTOR信号通路的损伤。mTOR信号通路除了在不同抗抑郁药中发挥作用外,与NMDA受体拮抗剂特别是氯胺酮有更紧密的联系。通过综述MDD与mTOR信号级联通路的关系及抗抑郁药对mTOR信号级联通路的作用原理,揭示mTOR信号通路在抑郁症中的特征及其对抗抑郁药的反应对开发新型抗抑郁药有重要的临床价值。 Postmortem examination reported that robust deficits in mTOR signaling pathway were investigated in prefrontal cortex of MDD patients. Nonetheless, mTOR signaling pathway was more associated with N-methyl-D-aspaTtate(NMDA) receptor antagonists, such as ketamine, be sides the aet t0n involved in effects of other antidepressants. The relationship between major depressive disordet(MDD) and the mammalian target of rapamycin(mTOR) signaling cascade pathway were studied in this article, which was implicated in rationale of roTOR signaling cascade pathway for antidepressants. It goes without saying that the characteristics of mTOR signa MDD and actions in response to antidepressants will exhibit great clinical significance opment of new antidepressants. g pathway in in the developmen of new antidepresssants.
