响应面法优化琼胶酶发酵培养基

Agarase Optimization of Fermentation Medium by Response Surface Method

摘要 [目的]优化海洋细菌Vibrio agarivorans-1产琼胶酶液体发酵培养基的最佳组分。[方法]首先通过单因素试验筛选出3个重要因子(琼脂、NaCl和酵母膏质量浓度),在单因素的基础上,通过Box-Behnken试验设计和响应面分析法确定了3个因子的最优发酵条件。[结果]最佳营养组分为:琼脂粉质量浓度为3.9 g/L,NaCl质量浓度为45.2 g/L,酵母膏质量浓度为12.9 g/L,此条件下酶活力达到7.589 U/g,比优化前提高了3.35倍。[结论]该研究对提高海洋细菌Vibrio agarivorans-1产琼胶酶具有重要意义。 [Objective] The components of liquid fermentation medium of marine microorganisms Vibrio agarivorans-1 producing agarase were optimized. [ Method ] Three important factors: the concentration of agar, NaC1 and yeast extract in liquid fermentation medium, were screened by single factor experiment. The optimum fermentation conditions were determined by three factors Box-Behnken experimental design and re- sponse surface analysis based on single factor experiment. [ Result] The experimental result indicated that the activity of agarase reached at 7. 602 U/g as the concentration of agar, NaCI and yeast extract was 3.9 g/L, 45.2 g/L and 12.9 g/L, respectively, which was 3.35 times higher than the previous. [ Conclusion] The research has significance for enhancing marine microorganisms Vibrio agarivorans-1 producing agarase.
