甜菜碱对酒精性肝病大鼠肝细胞色素P450 2E1基因和蛋白表达的影响

摘要 目的:观察甜菜碱对酒精性肝病(ALD)模型大鼠肝细胞色素P4502E1(CYP 2E1)基因和蛋白表达的影响。方法:建立大鼠酒精性肝病模型后,分别以甜菜碱不同剂量(200~800 mg/kg)治疗4周,观察肝组织的病理形态学改变;采用RT-PCR和免疫组化法检测CYP 2E1基因和蛋白表达水平,并分别用硫代巴比妥酸法(TBA法)和黄嘌吟氧化酶法测定肝组织丙二醛(MDA)的含量和超氧化物歧化酶(SOD)的活力。结果:甜菜碱高、中剂量组能显著减少肝脂质的沉积和炎症细胞的浸润;RT-PCR和免疫组化结果证明甜菜碱能显著抑制酒精性肝病CYP 2E1基因和蛋白的表达,同时肝内MDA和SOD含量恢复到接近正常。结论:甜菜碱能显著抑制酒精性肝病CYP 2E1基因和蛋白的表达,具有防止酒精性肝病的作用。 Objective: To survey the effects of betaine on the expressions of hepatocyte cytochrome P450 2E1 (CYP 2E1) gene and protein of the rats with alcoholic liver disease (ALD). Methods: After ALD rat model was es-tablished, they were treated by betaine in different dosages (200 to 800 mg/kg) for four weeks, the changes of liver tissue pathomorphology were observed; the levels of CYP 2E1 protein and gene were detected by RT-PCR and im-munohistochemical method, the contents of MDA and the activity of SOD were determined with TBA method and xanthine oxidase method. Results: High and moderate dosages of betaine could notably reduced liver lipid deposi-tion and the infiltration of inflammatory cells, RT-PCR and immunohistochemical results proved that betaine could remarkably inhibit the expressions of CYP 2E1 protein and gene in ALD patients, and recover the contents of MDA and SOD to normal. Conclusion: Betaine could effectively inhibit the expressions of CYP 2E1 gene and protein of ALD rats, which shows the function of preventing ALD.
