摘要
在单因素试验的基础上,采用响应面试验,优化了分光光度法测定金花茶总皂甙的显色反应条件.得到的最佳反应条件为:5%香草醛-冰醋酸溶液0.35 mL,高氯酸1.0 mL,反应温度80℃,反应时间25 min,测定波长540nm.在此条件下,人参皂甙Rg1在0~114.2 μg范围内与吸光度呈良好的线性关系,r=-0.9991.该方法的相对标准偏差为1.36%,稳定性相对标准偏差为2.08%,平均回收率为98.22%.
Based on single factor tests, the response surface method (RSM) was used to optimize the determination condition of total saponins in Camellia chrysantha by spectrophotometry. The optimal conditions were as follows: the amount of 5% vanillin-glacial acetic acid was 0.35 mL, the amount of perchloric acid was 1.0 mL, the reaaction temperature was 80℃, the reaction time was 25 rain and wavelength was 540 nm. Under the optimal conditions, the calibration curve was linear in the range of 0-114.2 μg, The relative coefficient was 0.9991. The relative standard deviations of precision and stability were 1.36% and 2.08%, respectively. The average recovery rate was 98.22%.
