摘要
目的探讨三七总皂苷(PNS)对老年大鼠学习能力、记忆力及海马神经元凋亡的影响。方法选择22月龄雌性Sprague-Dawley(SD)大鼠45只,随机分为低剂量组15只、高剂量组15只、老年对照组15只;另选择2月龄雌性SD大鼠15只作为青年对照组。低剂量组大鼠每天注射1次PNS,剂量为50μg/kg;高剂量组大鼠每天注射1次PNS,剂量为100μg/kg;青年对照组和老年对照组大鼠均注射等量0.9%氯化钠溶液。各组大鼠均连续注射30 d。对各组大鼠进行学习能力、记忆力测试,计算其正确反应率;采用TUNEL法检测海马CA1区神经元凋亡情况。结果青年对照组、低剂量组、高剂量组学习能力及记忆力测试正确反应率高于老年对照组,高剂量组学习能力及记忆力测试正确反应率高于低剂量组(P<0.05)。青年对照组、低剂量组、高剂量组海马CA1区TUNEL阳性细胞数低于老年对照组,高剂量组海马CA1区TUNEL阳性细胞数低于低剂量组(P<0.05)。结论 PNS能改善老年大鼠的学习能力和记忆力,减少海马神经元凋亡,大剂量应用时效果较好。
Objective To investigate the effects of panax notoginseng saponins( PNS) on learning ability,memory and neurons apoptosis of hippocampus in old rats. Methods A total of 45 22-month-old female Sprague-Dawley( SD) rats were chosen and randomly divided into groups A,B,C,each of 15 cases;while 15 2-month-old female SD rats were chosen as group D. group A given PNS,50 μg/kg,1 time per day;group B given PNS,100 μg/kg,1 time per day;groups C and D given equivalent 0. 9% sodium chloride solution. All groups treated for 30 days. Tests for learning ability and memory were carried out and rate of right action calculated;neurons apoptosis in the CA1 area of hippocampus was observed by TUNEL. Results The rates of right action in the tests for learning ability and memory of groups A,B,D were higher than those of group C,group B higher than those of group A. Positive cell counts of TUNEL in the CA1 area of hippocampus of groups A,B,D were lower than that of group C,group B lower than that of group A. Conclusion PNS can improve the learning ability and memory of old rats,reduce the neurons apoptosis of hippocampus,and higher dose of PNS has better effect.
