摘要
目的探讨粒细胞集落刺激因子(G-CSF)对脑出血(ICH)大鼠血管新生的影响及其作用机制。方法选取健康SD大鼠63只,随机分为假手术组、ICH组、治疗组,每组21只。制备ICH模型,治疗组大鼠造模1 h后腹腔注射重组人粒细胞集落刺激因子注射液60μg/kg,假手术组、ICH组大鼠经腹腔注射等体积的0.9%氯化钠溶液。比较3组大鼠于造模后6 h、24 h、48 h、72 h、7 d、14 d、21 d神经功能障碍评分、CD34阳性细胞数、血管内皮生长因子(VEGF)阳性细胞数。结果 ICH组大鼠造模后6 h、24 h、48 h、72 h、7 d、14 d、21 d神经功能障碍评分均低于假手术组,治疗组大鼠造模后24 h、48 h、72 h、7 d、14 d、21 d神经功能障碍评分均高于ICH组(P<0.05)。ICH组大鼠造模后6 h、24 h、48 h、72 h、7 d、14 d、21 d CD34阳性细胞数均多于假手术组,治疗组大鼠造模后6 h、24 h、48 h、72 h、7 d、14 d、21 d CD34阳性细胞数均多于ICH组(P<0.05)。ICH组大鼠造模后6 h、24 h、48 h、72h、7 d、14 d、21 d VEGF阳性细胞数均多于假手术组,治疗组大鼠造模后6 h、24 h、48 h、72 h、7 d、14 d、21 d VEGF阳性细胞数均多于ICH组(P<0.05)。结论 G-CSF可促进ICH大鼠损伤区域血管新生及神经功能恢复,其作用机制可能为上调CD34及VEGF的表达。
Objective To investigate the effect of granulocyte colony-stimulating factor( G-CSF) on angiogenesis in rats with intracerebral hemorrhage( ICH) and its mechanism. Methods A total of 63 healthy Spragne-Dawley rats were se-lected and randomly divided into groups A( given sham-operation),B( made ICH models),C( made ICH models),each of 21 cases. Then group C given rhG-CSF 60 μg/kg by intraperitoneal injection after 1 h of models made,while groups A and B given equal volume of 0. 9% sodium chloride solution. Neural function defection score,positive CD34 cell counts and positive VEGF cell counts were compared at 6 h,24 h,48 h,72 h,7 d,14 d,21 d after models made in the three groups. Results Neural function defection score of group B was significantly higher than that of group A,at 6 h,24 h,48 h,72 h,7 d,14 d,21 d after models made,respectively;and neural function defection score of group C was significantly higher than that of group B,at 24 h,48 h,72 h,7 d,14 d,21 d after models made,respectively(P﹤0. 05). Positive CD34 cell counts and positive VEGF cell counts of group B were significantly higher than those of group A,at 6 h,24 h,48 h,72 h,7 d,14 d, 21 d after models made;and positive CD34 cell counts and positive VEGF cell counts of group C were significantly higher than those of group B,at 6 h,24 h,48 h,72 h,7 d,14 d,21 d after models made. Conclusion G-CSF could promote the angiogenesis and improve the recovery of neural function,and its possible mechanism is upregulating the expression of CD34 and VEGF.
