调控水通道蛋白用于大鼠继发性脊髓损伤的实验效果研究 被引量:1

摘要 目的分析水通道蛋白在脊髓损伤后的变化规律,探讨水通道蛋白在脊髓损伤的治疗价值。方法通过建立大鼠Allen脊髓损伤动物模型,观察术后不同时间点AQP-4表达变化,图像分析仪定量分析AQP-4的变化。结果脊髓损伤后,AQP-4阳性表达显著增多,AQP-4及AQP-4mRNA呈高水平,显著高于正常对照组(P<0.05);与正常对照组比较,脊髓损伤组AQP-4染色积分光密度明显增加,在第3天达到峰值(P<0.05)。结论 AQP-4在脊髓损伤中具有重要作用,可能参与了水肿形成过程,AQP-4高表达可能是脊髓损伤性水肿的生物学机制之一。 Objective To analyze the changes of aquaporins in spinal cord injury and to explore the therapeutic value of aquaporins in spinal cord injury .Methods By establishing the animal model of Allen spinal cord injury in rats ,we observed the changes of AQP-4 expressions at different time points after surgery and we used image analyzer for making quantitative analysis of AQP-4 changes .Results After spinal cord injury ,AQP-4 positive expressions were increased significantly ,AQP-4 and AQP-4 mRNA were high ,they were significantly higher than those of the control group (P〈0 .05) .When compared with nor-mal control group ,AQP-4 staining score optical density was increased significantly in spinal cord injury group ,it reached the peak value in the 3th day ,the difference was significant (P〈0 .05) .Conclusion AQP-4 played an important role in spinal cord injury ;it might have been involved in the process of edema formation .AQP-4 over-expression might be one of the biological mechanisms of edema in spinal cord injury .
