摘要
目的观察不同浓度异丙酚静脉全身麻醉对大鼠海马各区c-fos、c-jun基因及蛋白表达的影响。方法 40只Wistar大鼠随机分成4组:空白对照组(A组)、小剂量异丙酚组(50 mg/kg,B组)、中等剂量异丙酚组(100 mg/kg,C组)和大剂量异丙酚组(150 mg/kg,D组)。采用反转录-聚合酶联反应和蛋白印迹方法检测大鼠海马各区c-fos、c-jun基因及蛋白的表达水平。结果 A组大鼠海马CA1区和DG区c-fos、c-jun基因表达水平为(0.32±0.06,0.27±0.05;0.37±0.07,0.28±0.07),c-fos、c-jun蛋白表达水平为(0.47±0.12,0.41±0.10;0.51±0.14,0.44±0.09);B组大鼠海马CA1区和DG区c-fos、c-jun基因表达水平为(0.54±0.11,0.41±0.10;0.61±0.12,0.44±0.12),c-fos、c-jun蛋白表达水平为(0.75±0.24,0.72±0.21;0.82±0.27,0.68±0.17);C组大鼠海马CA1区和DG区c-fos、c-jun基因表达水平为(0.87±0.23,0.75±0.18;0.92±0.27,0.72±0.21),c-fos、c-jun蛋白表达水平为(1.07±0.37,1.01±0.32;1.12±0.38,0.97±0.25);D组大鼠海马CA1区和DG区c-fos、c-jun基因表达水平为(1.22±0.30,0.96±0.24;1.31±0.34,0.95±0.28),c-fos、c-jun蛋白表达水平为(1.43±0.41,1.38±0.39;1.46±0.41,1.31±0.33)。大鼠海马CA1区和DG区c-fos、c-jun基因和蛋白的表达水平随着异丙酚麻醉药物作用浓度的增加显著性升高(P<0.05);海马CA3区未发现异丙酚对c-fos、c-jun基因和蛋白表达水平的影响。结论异丙酚可能通过影响c-fos、c-jun基因及蛋白的表达,从而发挥抑制中枢神经系统的作用,且此作用部位存在明显的差异性。
Objective To observe the effects of propofol anesthesia in different concentrations on the expression of c-fos,c-jun gene and protein in the different hippocampus regions of rat.Methods A total of 40 Wistar rats were randomly divided into 4 groups:control group(group A),low dose propofol group (50 mg/kg,group B),moderate dose propofol group(100 mg/kg,group C) and large dose propofol group (150 mg/kg,group D).The expression levels of c-fos,c-jun gene and protein in the rat hippocampus were detected by RT-PCR and Western-blot methods.Results The expression levels of c-fos,c-jun gene in the CA1 area and DG area of rat hippocampus of group A were (0.32 ± 0.06,0.27 ± 0.05 ; 0.37 ± 0.07,0.28 ± 0.07),and the expression levels of c-fos,c-jun protein were (0.47 ± 0.12,0.41 ± 0.10;0.51 ± 0.14,0.44 ± 0.09).The expression levels of c-fos,c-jun gene in the CA1 area and DG area of rat hippocampus of group B were (0.54 ± 0.11,0.41 ± 0.10 ; 0.61 ± 0.12,0.44 ± 0.12),and the expression levels of c-fos,c-jun protein were (0.75 ± 0.24,0.72 ± 0.21 ;0.82 ± 0.27,0.68 ± 0.17).The expression levels of c-fos,c-jun gene in the CA1 area and DG area of rat hippocampus of group C were(0.87 ± 0.23,0.75 ± 0.18 ; 0.92 ± 0.27,0.72 ± 0.21),and the expression levels of c-fos,c-jun protein were (1.07 ± 0.37,1.01 ± 0.32 ; 1.12 ± 0.38,0.97 ± 0.25).The expression levels of c-fos,c-jun gene in the CA1 area and DG area of rat hippocampus of group D were (1.22 ± 0.30,0.96 ± 0.24; 1.31 ± 0.34,0.95 ± 0.28),and the expression levels of c-fos,c-jun protein were (1.43 ± 0.41,1.38 ± 0.39 ; 1.46 ± 0.41,1.31 ± 0.33).As the concentration of propofol drug increased,the expression levels of c-fos,c-jun gene and protein in the CA1 area and DG area of rat hippocampus increased significantly(P 〈 0.05).There was no effects of propofol anesthesia on the expression of c-fos,c-jun gene and protein in the CA3 area of rat hippocampus.Conclusion Propofol may cause an inhibitory effect on the central nervous system by the expression of c-fos,c-jun gene and protein,and the sites of action are different.
