摘要
以花生幼叶为试材,对高通量DNA提取法、SDS简易法、TPS法以及TENP法4种不同的DNA提取方法进行比较和优化。结果表明,与其他3种改良方法比较,改良TPS法具有不需液氮磨样、提取速度快、试剂种类少、成本低、DNA溶解速度快、质量较好的优点,提取的DNA可用于花生SSR分析。
With the young leaves of peanut as experiment material,four different DNA extraction methods,the high-throughput DNA extraction method,the simple SDS method,TPS method and TENP method,were compared and optimized.The results showed that,compared with the other improved methods,the improved TPS method had the advantages of grinding samples with no liquid nitrogen,faster extraction rate,fewer kinds of reagent,lower cost,rapider DNA solution rate and better DNA quality.The DNA extracted through improved TPS method could be used for SSR analysis of peanut.
