摘要
目的测定春柴胡中柴胡皂苷a、d的含量。方法应用高效液相色谱(HPLC)法,色谱柱为DiamonsilC18(4.6 mm×250 mm,5μm),流动相为乙腈-水梯度洗脱,流速1mL·min-1,检测波长210 nm。结果柴胡皂苷a在0.4018~6.027μg(r=0.9990)范围内线性关系良好,平均回收率为91.19%,RSD为1.83%(n=6);柴胡皂苷d在0.5238~7.857μg(r=0.9992)范围内线性关系良好,平均回收率为92.76%,RSD为1.43%(n=6)。结论本研究建立的柴胡皂苷a、d的含量测定方法简便、结果准确。
Objective: To determine the saikosaponin a and d content in spring Bupleurum. Methods: HPLC was used. Column was Diamonsil C18(4.6 mm × 250 mm,5 μm),mobile phase was acetonitrile- water gradient elution,1mL·min- 1of flow velocity and210 nm of detection wavelength. Results: The saikosaponin a had a good linear relationship at range of 0. 4018 ~ 6. 027 μg(r = 0. 9990).The average recovery rate was 91. 19% and RSD was 1. 83%( n = 6); the saikosaponin d had a good linear relationship at range of0. 5238 ~ 7. 857μg(r 0. 9992). The average recovery rate was 92. 76% and RSD was 1. 43%(n = 6). Conclusion: The study establishes a determination method for saikosaponin a and d,which is simple and accurate.
