针刺对OLETF大鼠骨骼肌PKCζ/λ的影响 被引量:2

摘要 目的:观察针刺对OLETF大鼠骨骼肌PKCζ/λ的影响。方法:将16只雄性OLEFT大鼠随机分为2组,即模型组和针刺组,每组8只,8只SD大鼠作为空白组,用葡萄糖氧化酶法、ELISA、实时荧光定量PCR、Western blot等方法检测比较各组大鼠空腹血糖(FPG)、血浆胰岛素(FINS)、胰岛素敏感性指数(ISI)、血清C-肽(C-P)和骨骼肌PKCζ/λmRNA及其蛋白的表达。结果:针刺组的FPG、FINS、C-P较模型组显著降低(P<0.01),ISI较模型组显著升高(P<0.01),模型组的PKCζmRNA和PKCλmRNA表达较空白组表达显著降低(P<0.01)。针刺组PKCζmRNA表达较模型组显著升高(P<0.01),针刺组大鼠骨骼肌phospho-PKCζ/λ蛋白表达较空白组升高(P<0.05)。结论:针刺可能通过对骨骼肌phospho-PKCζ/λ转录水平的调节改善大鼠的胰岛素抵抗状态,调节转录表达的同时可能刺激了相应蛋白的表达,使phospho-PKCζ/λ蛋白表达有一定的提升。 Objective: To observe the curative effects of acupuncture on skeletal muscle PKCζ/λ of OLETF rats. Methods: 16 male OLEFT rats were divided randomly into two groups: the model group and the acupuncture group, 8 rats each group, and 8 SD rats were used as blank group. The fasting blood-glucose (FBG), fasting plasma insulin (FINS), insulin sensitivity index (ISI), Serum c-peptide(C-P), PKCζ/λ . mRNA and protein expression of skeletal muscle from each group were observed after treatment by Glucose oxidase method, ELISA, Real-time PCR and Western blot. Results: Compared with the model group, the FPG, FINS, C-P of the acupuncture group declined dramatically (P〈0.01), and ISI of the acupuncture group increased significantly (P〈0.01). Compared with the blank group, PKCζ mRNA and PKCλ mRNA expression of the model group declined evidently (P〈0.01). Compared with the model group, the PKCζ mRNA of the acupuncture group increased significantly (P〈0.01). Compared with the blank group, the PKCζ/λ protein expression of the acupuncture group rose significantly (P〈0.05). Conclusion: Acupuncture may improve the condition of insulin resistance in rats by regulating transcription of skeletal muscle phospho-PKCζ/λ , and while regulating the transcription expression, it may stimulate the corresponding protein expression, making the phospho-PKCζ/λ protein expression improve.
