白芍在配方颗粒冲剂与传统饮片煎剂薄层色谱鉴别及芍药苷含量比较 被引量:11

Comparing study on TLC identification and paeoniflorin content between granules and decoction of debark peony root

摘要 目的:通过白芍在配方颗粒冲剂与传统饮片煎剂薄层色谱鉴别及芍药苷含量比较,探讨中药配方颗粒在中药复方破癥逐瘀方中应用的可行性。方法:用薄层色谱法(TLC)鉴别处方中的白芍。采用高效液相色谱法(HPLC)测定破癥逐瘀方中芍药苷的含量:色谱柱:Lichrospher C18(200mm×4.6mm,5μm);流动相:乙腈∶0.1%磷酸溶液(14∶86);检测波长:230nm;柱温:35℃;流速:1.0mL/min。结果:在TLC上可检出白芍药材的特征斑点并且无干扰。TLC法测定中,在此色谱条件下,芍药苷在0.2704-2.7040μg范围内,进样量与峰面积积分值呈良好的线性关系(R=0.999)。结论:白芍在破癥逐瘀方中配方颗粒汤剂与传统饮片汤剂薄层色谱鉴别及芍药苷含量比较无显著性差异,提示其成分基本相同。 Objective: In order to discuss the feasibility of granules used in Chinese herbal compound-Pozheng Zhuyu Formula, this study took Pozheng Zhuyu Formula as an experimental object, and the paeoniflorin content in granules and decoction of Pozheng Zhuyu Formula was tested. Methods: Thin layer chromatography (TLC) was used to identify debark peony root. High performance liquid chromatography (HPLC) was used to test paeoniflorin content in Pozheng Zhuyu Formula. The separation of HPLC was performed on a Lichrospher C18 column (200mm×4.6mm, 5μm) with acetonitrile-0.1% phosphoric acid (14 : 86) as the mobile phase in a gradient elution at a flow rate of 1.0mL/min. The detect wavelength was set at 230nm, and the column temperature was set at 35℃. Results: The characters of debark peony root could be detected by TLC. Under the condition of chromatography, the paeoniflorin content in the range of 0.2704-2.70401ag. Sample quantity had a good linear relationship with peak area integral value (r=0.999, n=6). Conclusion: The TLC identification of debark peony root and paeoniflorin content in the granules and decoction of Pozheng Zhuyu Formula has no significant difference, which shows that the composition of granules and decoction is the same.
