丹参药材蛋白质提取工艺的优选研究 被引量:2

摘要 目的:优选丹参药材可溶性蛋白质的最佳提取工艺,为探讨丹参寒热药性与蛋白质含量的相关性提供实验依据。方法:对丹参药材蛋白质提取条件进行单因素考察,并采用L9(34)正交试验设计法,采用考马斯亮蓝染色法,以丹参药材可溶性蛋白质含量为指标,考察料液比、pH值、提取温度、提取时间四个因素对可溶性蛋白质提取率的影响,从而确定丹参药材蛋白质的最佳提取工艺。结果:建立了丹参药材蛋白质的最佳提取工艺:药材粉末中加入30倍量pH值为10的蒸馏水,在60℃条件下超声提取50 min。结论:优化的提取工艺稳定、合理、可行,能正确反映丹参药材中的蛋白质含量。 Objective:To optimize the extraction technology of protein in Salvia miltiorrhiza and to provide experimental basis for defining the relevance of traditional Chinese drugs property and protein content.Method:The optimized extraction technology was investigated by using the orthogonal test with the content of protein as the index determined by the method of coomassie brilliant blue staining. The effect of solid-liquid ratio,pH,temperature and time on the extraction rate were investigated. Results:The best extracting condition of protein in Salvia miltiorrhiza was established,which is the powder of Salvia miltiorrhiza was extracted for50 minutes with 30 times amount of water(pH 10) by ultrasonic at the temperature of 60 ℃. Conclusion:The optimum extraction condition was stable and reliable,and could reflect the protein content of Salvia miltiorrhiza.
