参柏复合乳质量标准研究

摘要 目的建立参柏复合乳的质量控制方法。方法采用薄层色谱法对参柏复合乳中主要成分关黄柏和苦参进行定性鉴别。采用高效液相色谱法测定盐酸小檗碱的含量。色谱条件:色谱柱为DiamonsilC18柱(250mm×4.6mm,5μm),流动相为乙腈-0.1%磷酸溶液(50:50)(每100ml加十二烷基磺酸钠0.1g),检测波长为265nm,流速1.2ml/min,柱温30℃。结果在薄层色谱中均能检出关黄柏和苦参药味的特征斑点。盐酸小檗碱进样量在0.122—1.22μg范围内与峰面积积分值线性关系良好(r=0.9998),平均加样回收率为96.17%,RSD=0.41%(n:6)。结论该方法操作简单,专属性强,准确可靠,重复性好,可用于参柏复合乳的质量控制。 Objective To establish the controlling methods of Shenbai Mixed Milk. Melhods To make qualitative identification of the main components-pheUodendron and sophora flavescens in Shenbai Mixed Milk by the method of TLC, measured the contents of berberine by HPLC. Chromatographic conditions: The chromatographic column is Diamonsil C18 (250 mm× 4.6 mm, 5 ×m), the mobile phase was acetonitrile -0.1% phosphoric acid (50: 50) (To add sodium dodecyl sulfonate 0. 1 g per 100ml), thedetection wavelength was 265 nm,the velocity was 1.2ml/min, the column temperature was 30℃. Results The features of phellodendron and sophora flavescens can be examined by TLC. The injection volume of berberine was between 0. 122 - 1.22 μg, it had a good relationship with the linear relationship of integral value of peak area ( r = 0. 999 8 ) , the averagerecovery rate is 96.17%, RSD = 0.41% ( n = 6). Conclusions This method can be easily operated, strongly specified, it is accurate and it has good reproducibility. This method can be applied to control the quality of Shenbai Mixed Milk.
