摘要
目的检测类风湿关节炎(RA)患者外周血CCR6+CD4+T细胞芳香烃受体(AhR)水平,探讨AhR在RA发病机制中的意义。方法采用流式细胞术测定33例RA患者及14例健康对照者外周血CCR6+CD4+T细胞中AhR阳性细胞比例,分析其与RA患者临床指标的关系。结果 1)RA患者组外周血单个核细胞(PBMCs)中CCR6+CD4+T细胞占(10.75±2.92)%,高于健康对照组(4.96±2.28)%,差异有统计学意义(t=6.581,P<0.01)。2)RA患者组内外周血CCR6+CD4+T细胞中AhR阳性细胞所占比例(46.02±14.68)%,高于CCR6-CD4+T细胞(21.62±6.94)%和PBMCs(34.22±10.33)%中AhR阳性细胞占比,差异均有统计学意义(z=-6.432,P<0.01;t=-3.776,P<0.01)。3)RA患者组外周血CCR6+CD4+T细胞中AhR阳性细胞所占比例(46.02±14.68)%,高于健康对照组(23.18±5.62)%,差异有统计学意义(z=-4.909,P<0.01)。4)相关性分析:RA患者组外周血CCR6+CD4+T细胞中AhR阳性细胞所占比例与血沉(ESR)、C反应蛋白(CRP)、抗环瓜氨酸肽(抗-CCP)抗体、DAS28评分、疾病病程均无相关性(P均>0.05)。结论 AhR阳性细胞在RA患者外周血CCR6+CD4+T细胞中比例高,可能参与了RA发病,但其水平高低不能反映RA疾病活动程度。
To explore the significance of aryl hydrocarbon receptor (AhR) positive eel1 expression in rheumatoid arthritis (RA), flow cytometry (FCM) was applied to detect the percentages of CCR6+CD4+T cells and AhR positive eells in peripheral blood mononuclear cells (PBMCs) of 33 cases with RA and 14 healthy controls. The detailed clinical data of the RA patients were recorded and the disease activity score (DAS) 28 was also calculated. Then, the relationship between percentage of AhR positive cells and DAS28 score was analyzed. Data showed that the percentage of CCR6+ CD4+T ceils in PBMCs from RA patients was significantly higher than that from healthy controls [(10.75±2.92)% vs (4.96±2.28)%, P〈0.01]. In peripheral blood of RA patients, was significantly higher than that in CCR6-CD4+T cells and tile percentage of AhR positive cells in CCR6+CD4+T cells PBMCs [(46.02±14.68)% vs (21.62±6.94)%, P〈0.01; (46.02± 14.68)% vs (34.22±10.33)%, P〈0.01, respectively]. Furthermore, the percentage of AhR positive cells in CCR6+CD4+T cells from RA patients was significantly higher than that from healthy controls [(46.02±14.68)% vs (23.18±5.62)%, P〈 0.01]. However, there was no significant correlation between DAS28 score and the percentage of AhR positive cells in CCR6+CD4+T cells. The results indicate that AhR positive cells in peripheral blood CCR6+CD4+T cells may be involved in development of rheumatoid arthritis, hut does not reflect disease activity.
