巴西橡胶树FCA基因的克隆及功能分析 被引量:2

摘要 FCA基因是植物自主开花途径中的重要调控基因,已被证实是促进植物开花的保守基因。研究通过巴西橡胶树甲基化文库获得橡胶树HbFCA基因一段434 bp的保守cDNA序列,应用RACE技术,成功克隆得到橡胶树的HbFCA基因。进一步通过构建HbFCA真核过表达载体pXCS-HbFCA-HAStrep,并转化拟南芥发现,转基因植株35 d后已经抽薹开花,野生型对照未开花,HbFCA的过表达能显著促进拟南芥开花。 Flowering control locus A(FCA) is an important regulation gene in plant flowering autonomous pathway,it has been proved to be the conservative gene for promoting flowering.In this study,a conserved fragment with 434 bp was obtained from the methylation filtration library in Hevea brasiliensis,based on which the HbFCA gene was cloned successfully by RACE technique.The eukaryotic overexpression vector pXCS-HbFCA-HAStrep was constructed and transformed into Arabidopsis thaliana.The results showed that the transgenic plants flowered after 35 days planting,but the wild type didn't bloom,which revealed that the overexpression of HbFCA could promote flowering in Arabidopsis significantly.
