超临界CO_2萃取川西老鹳草中鞣质工艺优选 被引量:3

Optimization of extraction process for tannins from Geranium orientali-tibeticum by supercritical CO_2 method

摘要 该文利用超临界CO2萃取装置对川西老鹳草中鞣质的提取工艺条件进行优化。采用磷钼钨酸-干酪素比色法测定鞣质的含量,以萃取压力、温度和时间为考察因素,以川西老鹳草萃取物中的鞣质含量为评价指标,通过正交试验对工艺条件进行优选。试验所得最佳工艺条件为萃取压力25 MPa,萃取温度50℃,萃取1.5 h。提取物中鞣质质量分数为12.91 mg·g-1,浸膏产率为3.67%。该比色法可以作为川西老鹳草鞣质的含量测定的方法,优化后的工艺稳定可行,提取率高,可为川西老鹳草中鞣质的提取工艺设计提供理论依据。 In order to optimize extraction process conditions of tannins from Geranium orientali-tibeticum by supercritical CO2,the content of tannins was determined by phosphomolybdium tungsten acid-casein reaction,with extraction pressure,extraction temperature and extraction time as factors,the content of tannins from extract of G.orientali-tibeticum as index,technology conditions were optimized by orthogonal test.Optimum technology conditions were as follows:extraction pressure was 25 MPa,extraction temperature was 50 ℃,extracted 1.5 h.The content of tannins in extract was 12.91 mg·g- 1,extract rate was 3.67%.The method established could be used for assay the contents of tannin in G.orientali-tibeticum.The circulated extraction was an effective extraction process that was stable and feasible,and that provides a way of the extraction process conditions of tannin from G.orientali-tibeticum.
