摘要
目的:通过检测磷酸化p38 MAPK(p-p38MAPK)在小鼠卵泡发育过程中的表达,为研究丝裂原活化蛋白激酶(MAPKs)信号通路在卵泡生长、发育及成熟过程中的作用提供理论基础。方法:随机选取健康成年小鼠60只,按雌雄2∶1的比例合笼繁殖新生小鼠。取3、5、7、11及21天雌性健康小鼠(各30只)的卵巢组织,采用免疫组化SP法检测不同年龄段小鼠卵巢组织中p-p38MAPK蛋白的表达水平。结果:p-p38MAPK蛋白在各年龄段小鼠不同级别卵泡的卵母细胞胞浆中均有表达,在始基卵泡中表达最强,与各级卵泡相比,差异均有统计学意义(P<0.05);初级卵泡与次级卵泡比较,差异无统计学意义(P>0.05);初级卵泡、次级卵泡与窦状卵泡相比,差异均有统计学意义(P<0.05)。该蛋白在各级卵泡颗粒细胞中也均表达,始基卵泡与各级卵泡相比,差异均有统计学意义(P<0.05);初级卵泡与次级卵泡相比差异有统计学意义(P<0.05),而与窦状卵泡相比差异无统计学意义(P>0.05);在次级卵泡中表达最强,与各级卵泡相比差异均有统计学意义(P<0.05)。不同年龄段小鼠卵巢中,21天小鼠卵母细胞p-p38MAPK蛋白表达强度分别与3天和7天小鼠进行比较,差异均有统计学意义(P<0.05);3、5、7及11天各组相比,差异均无统计学意义(P>0.05)。各年龄组颗粒细胞p-p38MAPK蛋白表达强度相比,差异均有统计学意义(P<0.05)。结论:p-p38MAPK蛋白在小鼠卵泡发育过程中具有时空特异性,p-p38MAPK通路可能在卵泡发育过程中起重要的调控作用。
Objective: To provide a theoretical basis for researching the effect of mitogen -activated protein kinase (MAPK) sig- nal pathway in follicular growth, development and maturity by detecting the expression of phosphorylated p38 MAPK ( p -p38 MAPK) during development of ovarian follicle in mice. Methods: Sixty health adult mice were randomly selected and neonatal mice were obtained after mating according to the ratio of 2: 1. The ovarian tissue samples were obtained from healthy female mice at 3, 5, 7, 11 and 21 days (30 mice in each time point) ; immunohistochemical SP method was used to detect the expression levels of p -p38 MAPK protein in ovarian tis- sue of mice in different age groups. Results: p - p38 MAPK protein expressed in cytoplasm of oocytes in follicles of different degrees in mice in different age groups, the expression level of p - p38 MAPK protein in primordial follicles was the highest, compared with the other follicles of different degrees, there were statistically significant differences ( P 〈 0.05 ) ; but there was no statistically significant difference between primary follicles and secondary follicles ( P 〉 0. 05 ), there were statistically significant differences between primary follicles, secondary folli- cles and antral follicles ( P 〈 0. 05) . p - p38 MAPK protein expressed in granulosa cells in follicles of different degrees, there were statistically significant differences between primordial follicles and the other follicles of different degrees ( P 〈 0. 05 ), there was also statistically significant difference between primary follicles and secondary follicles (P 〈 0. 05), but there was no statistically significant difference between primary follicles, secondary follicles and antral follicles ( P 〉 0.05 ) ; the expression level of p - p38 MAPK protein in secondary follicles was the highest, compared with the other follicles of different degrees, there were statistically significant differences (P 〈 0.05 ) . Among the mice in different age groups, there were statistically significant differences in expression level of p -p38 MAPK protein between 21 - day mice and 3 - day mice, 7 - day mice, respectively ( P 〈 0. 05 ), but there was no statistically significant difference between 3 - day mice, 5 - day mice, 7 - day mice and 11 - day mice ( P 〉 0. 05) ; there was statistically significant difference in expression level of p, p38 MAPK protein in granulosa cells among mice of different age groups ( P 〈 0. 05 ) . Conclusion: p - p38 MAPK protein has spatio - temporal specificity dUring, development of ovarian follicle in mice, p - p38 MAPK signal pathway may play an important regulating role during follicular development.
