醒脾解郁方对抑郁模型大鼠学习记忆行为及炎性因子水平的影响 被引量:5

摘要 目的探讨醒脾解郁方对抑郁模型大鼠的学习记忆行为和炎性因子水平的影响。方法将SD大鼠随机分为正常组、模型组、舍曲林组、中药组,采用21天慢性不可预见性温和应激建立大鼠抑郁模型,以Morris水迷宫实验评价大鼠的空间学习记忆能力,比较血清、海马炎性因子水平的变化。结果与正常组比较,模型组糖水偏爱率明显下降,水迷宫逃避潜伏期明显延长,血清、海马IL-1β、IL-6、TNF-α含量显著增加,差异均有统计学意义(P<0.05)。与模型组比较,在改善空间学习记忆方面,舍曲林组未见明显作用(P>0.05),中药组则表现出显著的改善作用(P<0.05);与模型组比较,中药组对血清及海马IL-1β、IL-6、TNF-α均有显著逆转作用(P<0.05),舍曲林组对血清、海马IL-6、TNF-α和海马IL-1β有显著改善作用(P<0.05),而对血清IL-1β无明显降低作用(P>0.05)。结论醒脾解郁方对改善抑郁大鼠学习记忆的作用优于舍曲林,可能与其降低抑郁模型大鼠血清、海马IL-1β、IL-6、TNF-α炎性因子的作用有关。 Objective To explore the effect of Xingpi Jieyu Decoction on learning-memory behavior and inflammatory factors level in depression rats. Methods SD rats were randomly divided into a normal group, a model group, a sertraline group and a TCM group. The chronic unpredictable mild stress for 21 days was adopted to establish the rat model of depression, spatial learning and memory ability of rats was evaluated by using Morris water maze, and the change of serum and hippocampus inflammatory cytokines was compared. Results Compared with the normal group, the preference for sugar in the model group was significantly decreased, the water maze escape latency was apparently increased and the content of IL-1 , IL-6 and TNF-α in serum and hippocampus was increased significantly ( P 〈 0. 05 ) ; Compared with the model group, in terms of improving spatial learning and memory, sertraline group had no difference ( P 〉 0.05 ) , TCM group had significant reverse effect on the content of IL-1 β, IL-6 and TNF-α in serum and hippocampus (P 〈 0. 05), while the sertraline group significantly improved the IL-6 and TNF-α in serum and hippocampus, as well as hippocampus IL-1β (P 〈 0. 05), nor the serum IL-β ( P 〉 0. 05 ). Conclusion Xingpi Jieyu Decoction has the better effect on improving learning and memory ability in depression rats than sertraline, which might be related to reduce the inflammatory factors level, such as IL-1β. IL-6 and TNF-α. in serum and hippocampus.
