摘要
为了测定运输应激前后夏南牛血清中促肾上腺皮质激素、皮质醇、白介素-6、肿瘤坏死因子-α水平,观察其变化规律,试验选用8头体重相近的8月龄健康夏南牛,分别于运输前1,0小时,运输中2,4,7,10,20小时,运输后15,30小时及2,3,7,15,21天采集血液,分离血清,用放射免疫法测定各血清样品中血浆促肾上腺皮质激素、皮质醇、白细胞介素6、肿瘤坏死因子-α含量。结果表明:与运输前1小时相比,促肾上腺皮质激素在运输中2,4,10,20小时及运输后15小时极显著升高(P<0.01),在运输后30小时恢复到正常水平;皮质醇在运输过程中显著升高(P<0.05),在运输后2天恢复到正常水平;白介素-6在运输中4小时显著升高(P<0.05),在运输后15小时极显著升高(P<0.01),在运输后2天恢复到正常水平;肿瘤坏死因子-α在运输中6小时极显著升高(P<0.01),在运输后30小时恢复到正常水平。说明短时间运输应激会刺激免疫细胞因子促肾上腺皮质激素、皮质醇、白细胞介素6、肿瘤坏死因子-α水平增加,而过长时间的运输则会造成免疫抑制。
To determine the levels of serum adrenocorticotropic hormone (ACTH), cortisol (COR) , interleukin -6 (IL -6 ) and tumor necrosis factor - alpha ( TNF - ct) in Xianan cattle before and after transportation stress. Eight healthy eight - month - old xianan cattle with similar weight were selected, and then their blood samples were collected to isolate serum at 0 h and 1 h before transportation; 2 h,4 h, 7 h, 10 h and 20 h during transportation; 15 h, 30 h, 2 d, 3 d, 7 d, 15 d and 21 d after transportation, respectively. The levels of plasma ACTH, COR, IL - 6 and TNF -α from each serum sample were determined using radioimmunoassay. The results showed that, compared with 1 hour before transportation, the levels of ACTH were significantly increased (P 〈0.01 ) at 2 h, 4 h, 10 h and 20 h during transportation, and 15 h after transportation, and then retumed to normal level at 30 h after transportation. The levels of COR during the whole transport process were significantly increased ( P 〈 0.05 ), then slowly returned to normal level at 2 d after transportation. The levels of IL - 6 were significantly increased ( P 〈 0.05 ) at 4 h during transportation, and were extremely significantly increased ( P 〈 0.01 ) at 15 h after transportation, then returned to normal levels at 2 d after transportation. The levels of TNF - α were significantly increased (P 〈0.01 ) at 6 b during transportation, then returned to normal level at 30 h after transportation. The results indicate that short - term transportation stress can stimulate the levels of ACTH, COR, IL- 6, and TNF-α as immune eytokines, but transportation over a long period of time will cause immunesuppression.
