九龙牦牛H-FABP与ADD1基因的表达谱研究 被引量:4

Study on the expression profiles of H-FABP and ADD1 genes in Jiulong yaks

摘要 为了研究心脏型脂肪酸结合蛋白(Heart fatty acid—binding protein,H—FABP)与脂肪细胞决定和分化因子1(Adipocyte determination and differentiation factor-1,ADD1)在九龙牦牛肉质形成中的作用,试验以九龙牦牛为研究对象,利用半定量RT—PCR检测其在九龙牦牛不同组织和不同发育阶段的表达差异,获得其组织表达和时序表达谱。结果表明:检测到H—FABP基因在九龙牦牛的心脏、脾脏、背最长肌和脂肪组织中表达,在肝脏和肾脏中未表达;ADDl基因仅在九龙牦牛的背最长肌和脂肪中表达,在心脏、肝脏、脾脏和肾脏中均未表达。时序表达谱分析结果显示,H—FABP与ADD1基因在0.5岁牦牛背最长肌中的表达丰度显著高于3.5~5.5岁和9岁以上的牦牛(P〈0.05),3.5—5.5岁牦牛背最长肌中的表达丰度显著高于9岁以上的牦牛。 To study the functions of heart fatty acid - binding protein ( H - FABP) and adipocyte determination and differentiation factor 1 ( ADD1 ) on meat quality formation in Jiulong yaks ( Bos grunniens), the tissue and temporal expression profiling of H - FABP and ADDI genes of Jiulong yaks were obtained using semi - quantitative RT - PCR to detect the expression differences in different tissues and different develop- mental stages in Jiulong yaks as research object. The results showed that H - FABP gene expressions could be detected in heart, spleen, longis- simus muscle and fat tissues in Jiulong yaks, but not in the liver and kidney. The ADD1 gene could only be expressed in longissimus muscle and fat tissues in Jiulong yaks, but not in the heart,liver, spleen, and kidney. The results of temporal expression profile analysis showed that the expression abundances of H - FABP and ADD1 genes in longissium muscle in the yaks at 0.5 year of age were significantly higher than those in the yaks at 3.5 to 5.5 years of age and over 9 years of age ( P 〈 0.05 ), and the expression abundances of H - FABP and ADD1 genes in longissium muscle in the yaks at 3.5 to 5.5 years of age were significantly higher than those in the yaks at over 9 years of age.
