GFI1B在急性髓系白血病M6和M7亚型中的表达及临床意义

摘要 目的探讨急性髓系白血病中急性红白血病(M6)和急性巨核细胞白血病(M7)亚型细胞患者独立生长因子1B(GFI1B)的异常表达及其临床意义。方法选择急性髓系白血病系初治住院患者14例(急性髓系白血病M6型10例,M7型4例)作为观察组,GFI1B表达正常者10例对照组。采用实时荧光定量PCR法检测观察组治疗前后GFI1B表达水平并与对照组比较。结果治疗前观察组GFI1B表达水平明显高于对照组,差异有统计学意义(P<0.05);治疗后观察组M6患者中缓解6例,GFI1B平均表达水平明显低于治疗前,差异有统计学意义(P<0.05),略高于对照组,但差异无统计学意义(P>0.05);复发4例,GFI1B平均表达水平较治疗前比较差异无统计学意义(P>0.05),与缓解患者及对照组比较差异有统计学意义(P<0.05)。M7患者中3例未行化疗,1例化疗后未缓解,未继续监测其GFI1B mRNA表达水平。结论 GFI1B的表达水平反映M6和M7亚型的病情变化,对其诊断、治疗以及预后检测有着重要的意义。 Objective To investigate the expression of GFI1 B and its clinical significance in acute leukemia and acute megakaryocytic leukemia. Methods Selected patients with acute myeloid leukemia in 14 cases( 10 cases of type M6,4cases of type M7) as the observation group,and 10 normal controls. The GFI1 B expression of observation group before and after treatment detected by real-time fluorescence quantitative PCR,and compared with the control group. Results The GFI1 B levels of observation group before treatment were significantly higher than control group,the differences were statistically significant( P〈0. 05). GFI1 B levels of 6 cases of type M6 in complete remission after treatment,were significantly lower than that before treatment,the differences were statistically significant( P〈0. 05),higher than control group,There was no statistically significant( P〉0. 05); 4 cases of recurrence,GFI1 B levels compare with before treatment had no significant difference( P〉0. 05),with complete remission patients and control group,the differences were statistically significant( P〈0. 05). 3 cases of type M7 without chemotherapy,1 patients without remission after chemotherapy. Conclusion Changes in GFI1 B levels reflect the condition of the patient with acute leukemia and acute megakaryocytic leukemia,has an important significance to the diagnosis,treatment and prognosis of patients.
