摘要
目的探讨磷酸化信号蛋白p38(P-p38)和基质金属蛋白酶-7(matrix metalloproteinases-7,MMP-7)在乳腺癌组织及癌旁正常乳腺组织中的表达及相关性。方法采用免疫组化SP法检测80例乳腺癌组织和20例癌旁正常乳腺组织中P-p38和MMP-7的表达,分析二者表达与临床病理参数的关系及相关性;采用Western blot法检测20例发生淋巴结转移的乳腺癌组织(转移组)、20例未发生淋巴结转移的乳腺癌组织(未转移组)及20例癌旁乳腺组织(对照组)中P-p38和MMP-7的表达。结果免疫组化检测:P-p38和MMP-7在乳腺癌组织中的阳性率分别为57.50%和77.50%,明显高于癌旁乳腺组织(P<0.05)。P-p38表达与乳腺癌的临床分期及淋巴结转移有关(P<0.05)。MMP-7表达与乳腺癌的组织学分级和淋巴结转移有关(P<0.05)。Western blot法检测:P-p38和MMP-7在转移组中表达最高,在未转移组表达高于对照组。相关性分析显示,乳腺癌中P-p38和MMP-7表达呈正相关性(rs=0.324,P=0.004)。结论乳腺癌组织中存在P-p38过度活化和MMP-7高表达,二者呈正相关,提示P-p38和MMP-7在乳腺癌的发生、发展中起促进作用,二者可能存在某种协同关系。
Purpose To investigate the expression and correlation of phosphorylated p38 (P-p38) and matrix metalloproteinases-7 (MMP-7) in pericancerous tissues and breast carcinoma tissues. Methods The expressions of P-p38 and MMP-7 protein in 80 cases of breast carcinoma and 20 cases of normal breast tissue were examined using immunohistochemistry method, and the relationship between the expressions of P-p38 and MMP-7 and their clinical pathological features was analysed. The expressions of P-p38 and MMP- 7 protein were determined using Western blot method in 20 cases of breast cancer tissues with lymph node metastasis, 20 cases of breast cancer tissues without lymph node metastasis, and 20 cases of adjacent breast tissues. Results The immunohistochemical results showed the expressions of P-p38 and MMP-7 in breast carcinoma tissues were significantly higher than that in adjacent breast tissues (P 〈 0. 05). The expression of P-p38 was correlated with lymph node metastasis and clinical stage (P 〈 0. 05). The expression of MMP- 7 was correlated with lymph node metastasis and histological grade (P 〈 0. 05). Western blot results showed that the expressions of P- p38 and MMP-7 were higher in lymph node positive group than that in lymph node negative group and control group. The Spearman' s correlation test revealed that the P-p38 was significantly positively associated with MMP-7 in breast carcinoma specimens (rs = 0. 324, P = 0. 004). Conclusion Overexpressions of P-p38 and MMP-7 were positively correlated in breast carcinoma tissues. The results suggested that P-p38 and MM-7 may cooperatively promote the growth and development of breast carcinoma.
