摘要 目的探讨高迁移率蛋白B1(HMGB1)影响人永生化角质形成细胞(HaCaT)分泌维持黑素细胞生长相关细胞因子的作用。方法 ELISA法检测HMGB1在正常对照组和白癜风患者血清中的表达。免疫荧光和实时定量PCR检测HMGB1及其受体Toll样受体2(TLR2),Toll样受体4(TLR4)和糖基化终产物受体(RAGE)在HaCaT中的表达。ELISA检测白癜风患者血清对于HaCaT分泌碱性成纤维细胞生长因子(bFGF)、干细胞因子(SCF)、内皮素-1(ET-1)的影响。结果相比于对照组,HMGB1在白癜风患者血清中表达升高;HMGB1及其受体在HaCaT中有表达,HMGB1表达在细胞核内;白癜风患者血清培养能够显著降低bFGF,SCF,ET-1的表达。结论角质形成细胞能够在病理状态下(白癜风)释放HMGB1,HMGB1可以降低角质形成细胞分泌的三种细胞因子从而参与白癜风的病理进程。 Objective To investigate the impact of high-mobility group protein B1 ( HMGBI ) on three kinds of growth factors secreted by keratinocytes. Methods HMGB1 expression from serum of vitiligo patients and controls was examined by ELISA. HMGBI expression in HaCaT cells was measured by immunofluorescence and HMGB1 receptors (TLR2,TLR4, RAGE) were quantitated by Real-time PCR. BFGF, SCF, ET-1 expression were examined from supernatants of cultured HaCaT treated by serum of vitiligo patients. Results HMGBl expression level was elevated in vitiligo patients compared with control. HMGB1 and its receptors were ex- pressed in HaCaT cells and HMGBl was expressed in the nucleus. Serum of vitiligo patients reduced the bF- GF, SCF,ET-1 secretion by HaCaT cells. Conclusion HMGB1 could be released by keratinocytes in patho- logical conditions (vitiligo) and reduce the secretion of growth factors contributing to the pathological process of vitiligo.
