摘要
采用猪瘟病毒C株毒(CSFV C strain)免疫BALB/C小鼠,按照常规单克隆抗体制作方法构建并筛选出能稳定分泌抗猪瘟单克隆抗体杂交瘤细胞株4株,分别命名为1C9,1B11,3C8和3G9,经鉴定,4株单克隆抗体腹水效价达到104~106。交叉试验及特异性试验表明,所制备的McAb与鸽新城疫病毒、猪蓝耳病病毒、猪传染性胃肠炎病毒、鸡减蛋综合征病毒无交叉反应性,均完全针对猪瘟病毒(CSFV)抗原决定簇,具有高度特异性。抗CSFV McAb的成功制备,为进一步研究建立CSFV的快速诊断方法奠定了基础。
Four monoclonal antibodies designated as 1C9,1B11,3C8 and 3G9 against classical swine fever virus C strain(CSFV C strain) were generated by immunization of BALB/C mice with puriifed CSFV C strain. The titers of ascetic fluids of the four McAbs were up to 104~106 in indirect ELISA. The McAbs were only positive against CSFV, and negative against Newcastle disease virus of pigeon,porcine reproductive and respiratory syndrome virus,trans-missible gastroenteritis virus and egg drop syndrome virus. The preparation of the McAbs provided a basis to establish rapid diagnostic method of CSFV.
