白色假丝酵母菌感染对口腔上皮细胞人类β-防御素2的表达 被引量:1

摘要 目的探讨白色假丝酵母菌对人类β-防御素2(HBD-2)在口腔上皮细胞中表达的影响,为口腔白色假丝酵母菌感染的治疗提供参考依据。方法试验分成3组,分别为孢子、菌丝及灭活的白色假丝酵母菌组;将KB细胞与孢子相、菌丝相及灭活的白色假丝酵母菌培养3、6、12、24h后,采用ELISA试验检测各组KB细胞培养液中HBD-2的表达,重复检测3次,采用SPSS18.0软件进行分析。结果随着时间的延长,黏附KB细胞的白色假丝酵母菌的总数由(1.83±0.64)CFU/cm2下降至(0.18±0.09)CFU/cm2,但活力未发生改变,菌丝转换率由(28.16±5.14)%增高至(80.18±10.14)%;感染24h后孢子相、菌丝相及灭活的白色假丝酵母菌组HBD-2分别为(60.08±6.47)pg/ml、(81.52±8.20)pg/ml、(64.81±7.35)pg/ml,均高于对照组(P<0.05)。结论白色假丝酵母菌在特定时间内可使口腔上皮细胞上调HBD-2表达,菌丝进一步侵袭后,HBD-2的表达降低。 OBJECTIVE To explore the impact of Candida albicans infection on expression of human β-defensin 2in oral epithelial cells so as to provide guidance for treatment of oral C.albicans infection.METHODS The study was designed as three groups:the blastospores group,the hyphae group,and inactivated C.albicans group,then the expression of human β-defensin 2in the KB cell culture fluid was determined by using ELISA after the KB cells,blastospores,hyphae,and inactivated C.albicans were cultured for 3,6,12,or 24 hours,which was repeated for three times,and the statistical analysis was performed with the use of SPSS18.0 software.RESULTS With the extension of time,the counts of C.albicans adhered to KB cells decreased from(1.83±0.64)CFU/cm2 to(0.18±0.09)CFU/cm2,but the bacterial vitality did not change;the conversion rate of hyphae increased from(28.16±5.14)% to(80.18±10.14)%.After being infected for 24 hours,the expression of human β-defensin 2was(60.08±6.47)pg/ml in the blastospores group,(81.52±8.20)pg/ml in the hyphae group,(64.81±7.35)pg/ml in the inactivated C.albicans,higher than those of the control group(P0.05).CONCLUSION C.albicans can induce the expression of human β-defensin 2in oral epithelial cells in a certain period;with the further attack of hyphae,the expression of human β-defensin 2decreases.
