干细胞因子对小鼠附植前胚胎体外发育的影响

摘要 为优化胚胎体外培养体系,提高胚胎体外培养发育率和发育质量,以昆明小鼠体内受精胚、体外受精胚和孤雌激活胚为研究对象,研究不同浓度干细胞因子对小鼠不同来源胚胎体外发育的影响。结果表明:在KSOM培养液中,小鼠自然受精胚、体外受精胚和孤雌激活胚的卵裂率和桑椹胚率均无显著差异(P>0.05),但IVN胚和IVF胚的囊胚发育率及囊胚细胞总数显著高于孤雌激活胚;培养液中添加不同浓度的干细胞因子对小鼠自然受精胚和孤雌激活胚的卵裂率、桑椹胚率、囊胚发育率和囊胚细胞总数都没有显著的影响(P>0.05),但干细胞因子浓度为100ng·mL-1时,小鼠体外受精胚的桑椹胚率和囊胚发育率显著高于对照(P<0.05)。 Abstract:In order to optimize in vitro culture system of embryo to improve development rate and quality, taking in vivo fertilization embryo,in vitro fertilization embryo and parthenogenetic activation as research object, the effect of different concentrations of stem cell factor on embryo culture in vitro were studied. The results showed that the cleavage rate and morula rate had no significant difference in KSOM, but the blastocyst rate and total blastoeyst number derived from PA was less than IVN and IVF. Different concentrations of stem cell factor had no significant effect on cleavage rate,morula rate, blastocyst rate and total blastocyst number. When the concentration of stem cell factor was 100 ng·mL-1 ,the morula rate and blastocyst rate were significantly higher than CK.
