RP-HPLC法同时测定天智颗粒中天麻素、栀子苷、芦丁和黄芩苷 被引量:11

Simultaneous determination of gastordine, geniposide, rutin and baicalin in Tianzhi Granules by RP-HPLC

摘要 目的建立同时测定天智颗粒(天麻、钩藤、槐花、栀子、黄芩、石决明、桑寄生、首乌藤)中天麻素、栀子苷、芦丁、黄芩苷量的RP-HPLC方法。方法采用ZORBAX SB-C18色谱柱(4.6 mm×250 mm 5μm);以乙腈-0.05%的磷酸水溶液为流动相,梯度洗脱,体积流量1.0 mL/min;柱温25℃;检测波长为220 nm。结果在220 nm检测波长下所测成分均有较强的吸收。天麻素、栀子苷、芦丁、黄芩苷的进样量分别在0.090 8~0.908μg(r=0.999 9)、0.288 1~2.881μg(r=0.999 8)、0.304 4~3.044μg(r=0.999 6)、0.3751~3.751μg(r=0.999 7)范围内线性关系良好;平均加样回收率分别为98.9%、98.0%、99.0%、98.1%,RSD分别为1.3%、0.7%、0.9%、1.0%。结论同时检测4种成分的RP-HPLC法优于只测定了天麻素的天智颗粒现执行标准。 AIM To establish RP-HPLC method for simultaneously determining the contents of gastordine, ge- niposide, rutin and baicalin in Tianzhi Granules (Gastrodiae Rhizoma, Gardeniae Fructus, Sophorae Flos, Scutel- lariae Radix, etc. ). METHODS ZORBAX SB-C18 (4. 6 mm× 250 mm 5 microns) chromatographic column was used for analysis with acetonitrile and 0. 05% phosphoric acid solution as mobile phase in gradient elution man- ner, 1.0 mL/min volume flow rate, 25 ℃ column temperature, and 220 nm detection wavelength were adopted. RESULTS The measured constituents had a good absorption under 220 mm detection wavelength. And the exper- iments showed a good linearity in the range of 0. 090 8 - 0. 908 txg ( r = 0. 999 9) for gastrodia, 0. 288 1 - 2. 881 ixg (r =0. 999 8) for geniposide, 0. 304 4 -3.044 μg (r =0. 999 6) for rutin, 0. 375 1 -3.751 μg (r = 0. 999 7) for baicalin, respectively. The average recoveries were 98.9% , 98.0% , 99.0% , and 98. 1% respec- tively. The RSD was 1.3% , 0. 7% , 0. 9% , and 1.0% respectively. CONCLUSION The method of simultane- ous determination of four constituents is superior to the current method for Tianzhi Granules which is only used for the determination of gastrodine.
