摘要
通过KOH-甲醇、硫酸-甲醇的回流甲酯化法与GB28404-2012标准的甲酯化法进行对比,发现前者可以将南极磷虾油中的各种脂肪酸快速、有效地转化成脂肪酸甲酯,用毛细管气相色谱法进行测定,拟合标准曲线建立EPA和DHA的外标定量分析方法,同时与峰面积归一化法进行对比。该法对EPA、DHA的相对标准偏差分别为1.9%和4.7%,回收率分别为98.6%和98.3%;外标法的重现性好,准确度高。
The methyl method which firstly saponified with KOH-CH 3OH solution and then the fatty acids are methylated with H2SO4-CH3OH solution,and GB 28404-2012 standard methyl ester method were compared, found that the former can be in a variety of Antarctic krill oil fatty acids quickly and efficiently converted into fatty acid methyl by capillary gas chromatography measurement, quantitative analysis of the external standard method to establish the standard curve fitting EPA and DHA, while the peak area normalization method for comparison. The RSD values of the method for the determination of EPA and DHA were 1.9%and 4.7%, respectively. And the recoveries of EPA and DHA were 98.6 % and 98.3 %, respectively.This method has high recovery rate and accurate quantitative analysis results in determination of EPA and DHA in Antarctic krill oil.
