摘要
以角豆胶为碳源,从四川雅安魔芋种植地的土壤中筛选得到1株高产β-甘露聚糖酶的细菌,通过形态观察和BIOLOG生理生化试验初步鉴定为枯草芽孢杆菌,命名为MSJ-5。初步优化了该菌的产酶配方:魔芋粉3.5%,蛋白胨1.0%,酵母膏0.5%,起始pH为8.5。培养温度为32℃,220 r/min,培养32 h即达产酶最高峰,酶活为642 U/mL。菌株MSJ-5能利用简单廉价的碳源和氮源高产甘露聚糖酶,且发酵周期短,在工业化生产甘露聚糖酶中具有很大的应用潜力。
A β-mannanase producing strain was isolated from Sichuan konjac planting field with carob bean gum as carbon source. The strain selected was named MS J-5 and identified as Bacillus subtilis based on morphological observation, and BIOLOG system. Optimization of the enzyme production of strain MSJ-5 was carried out and when the formulation was 3.5% konjac powder, peptone 1%, yeast extract 0. 5%, initial pH 8.5, the culture temperature was 32 ℃, 220 r/min, 32 hours of training, enzyme activity of MS J-5 could reach 642 U/mL in the culture liquid of shaking flask. Strain MS J-5 could produce high yield of β-mannanase with simple carbon source and nitrogen source in short fermentation period, which showed a great application potential in the industrial production of β-mannanase.
