摘要
采用浅盘法分离采集太谷自由生活的线虫,经分离并脱水制片后,用微分干涉显微镜进行形态观察和数值测量,并结合LSU和SSU的序列比对鉴定该线虫。结果表明,其雌虫形态特征为热杀死后向腹部弯曲,头部6根长刚毛和4根短刚毛位于同一圈,背部牙齿位于亚腹部牙齿的前端,背部牙齿和亚腹部牙齿形状均呈三角形,大小相似;在亚腹中部有颈刚毛1根;无后阴子宫囊;尾部有1对刚毛;雌虫体长924-1204μm,a为22—28,b为5.0—6.0,c为16.2~21.2;c’为2.0—2.7;V为65%~67%;雄虫未发现。将其LSU和SSU序列测序后与NCBI中已有的Tripylinazhejiangensis序列比对,相似率分别达100%和99%。根据其形态学和序列特征将其鉴定为Tripylina zhejiangensis,此种为山西省首次报道。
The morphological and molecular characters were used to identify the nematodes found in Taigu county, Shanxi province, China. The nematode is characterized by having a triangular dorsal tooth, pointing towards the ventral side, anterior to two sub-ventral teeth, with dorsal and sub-ventral teeth similar in shape and size; one ventromedian setae in the cervical region; absence of post-uterine sac; and a pair of setae on the tail; female body length 924-1 204 μm, a = 22-28, b = 5.0-6.0, c = 16.2-21.2, c' = 2.0-2.7 and V = 65%-67%; male not found. When compared the molecular data of present species with the published sequences in the GenBank, there are 100% nucleotide similarity for D2D3 region of LSU and 99% nucleotide similarity for SSU with Tripylina zhejiangensis. So the nematode was identified as T. zhejiangensis and it was the first time to be reported in Shanxi province, China.
