摘要
目的了解金华市淡色库蚊对杀虫剂的抗性基因,分析蚊虫抗性形成的机制,为蚊虫防制提供依据。方法采用PCR技术检测,利用Vector序列分析软件进行比对分析。结果金华市淡色库蚊现场品系与敏感品系抗性相关基因中vssc基因序列差异较大,序列相似度仅有84.1%;Est-2基因、ace1基因和cyp6f1基因序列分别存在4个、2个和1个核苷酸的差异;NYD-GBE、NYD-MLC2基因序列无差异。通过将发生点突变的基因序列翻译成氨基酸序列再进行比对发现,Est-2基因、ace1基因和cyp6f1基因的点突变均为无意突变,不会导致氨基酸序列变化。结论本研究中检测的6个基因片段中,金华市淡色库蚊现场与敏感品系的氨基酸序列完全相同。
Objective To study the resistance gene of Culex pipiens pallens to insecticides and to analyze the mechanism of insect resistance, so as to provide basis for reasonable use of insecticide. Methods The PCR technology was used to detect the related resistance genes in the field strains and susceptible strains of Culex pipiens paUens. Then the vector sequence analysis software was used for comparative analysis of the two categories of strains. Results The vssc gene sequences showed large differences between the field strains and susceptible strains of Culex pipiens pallens from Jinhua, with only 84.1% gene sequence similarity. While the Est -2 gene, acel gene and cyp6f1 gene in the field strains and susceptible strains showed difference of 4 nucleotides, 2 nucleotides and 1 nucleotide respectively. The gene sequence of NYD - GBE gene and NYD - MLC2 gene had no difference. Through comparing the amino acid sequences which were translated from the point mutation gene sequences, it was found that the point mutations of the Est -2 gene, acel gene and cyp6f1 gene were nonsense mutations, leading to no changes in amino acid sequence. Conclusion The field strains and susceptible strains of Culex pipiens pallens showed the identical amino acid sequence in Jinhua through the study on six gene fragments.
