脂肪源性干细胞移植对糖尿病周围神经病大鼠坐骨神经TNF-α和MBP表达的影响 被引量:1

Effects of adipose derived stem cells transplantation on the expression of TNF-α and MBP in sciatic nerve of rats with diabetic peripheral neuropathy

摘要 目的研究脂肪源性干细胞(ADSC)移植对糖尿病周围神经病(DPN)大鼠坐骨神经中肿瘤坏死因子-α(TNF-α)和髓磷脂碱性蛋白(MBP)表达的影响。方法大鼠随机分为正常组、模型组和ADSC组。链脲佐菌素腹腔注射制备实验DPN大鼠模型,ADSC组尾静脉注射ADSC细胞悬液,造模后14 d检测各组血糖及坐骨神经传导速度,实时荧光定量PCR检测坐骨神经TNF-α和MBP mRNA表达。结果与正常组比较,模型组和ADSC组血糖显著增高,神经传导速度显著降低,坐骨神经TNF-α和MBP mRNA表达显著上调。与模型组比较,ADSC组神经传导速度显著增高,坐骨神经TNF-α和MBP mRNA表达显著下降。结论脂肪源性干细胞下调糖尿病周围神经病坐骨神经TNF-α和MBP mRNA表达。 Objective To investigate the effects of transplantation of adipose-derived stem cells(ADSC) on the expression of tumor necrosis factor-α(TNF-α) and myelin basic protein(MBP) in the sciatic nerve of rats with diabetic peripheral neuropathy(DPN). Methods The rats were randomly divided into normal group, model group, ADSC group. The rat model of diabetic peripheral neuropathy was established by injection of streptozotocin peritoneally. ADSC suspension was infused into the caudal vein in ADSC group. 14 days after modeling, the blood sugar level and conduction velocity of sciatic nerve were measured, the mRNA expression of TNF-α and MBP in the sciatic nerve was detected by real time-PCR.Results Compared with normal group, the blood sugar level was increased and the conduction velocity of sciatic nerve was decreased in model group and ADSC group( 〈0.05), the expression levels of TNF-α mRNA and MBP mRNA in sciatic nerve were increased( 〈0.05). The conduction velocity was higher( 〈0.05), and the expression levels of TNF-α mRNA and MBP mRNA in sciatic nerve were decreased( 〈0.05) in ADSC group than in the model group. Conclusion ADSC could downregulate the expression levels of TNF-α mRNA and MBP mRNA in sciatic nerve of rats with diabetic peripheral neuropathy.
