复方五花血藤对鼠视网膜缺血再灌注损伤的保护作用 被引量:4

摘要 目的探讨复方五花血藤对大鼠视网膜缺血再灌注损伤的保护作用。方法采用健康成年SD大鼠90只,随机分为正常对照组、模型组和治疗组,其中模型组和治疗组建立视网膜缺血再灌注模型,治疗组于造模前连续3 d给予复方五花血藤灌胃,每天2次(总剂量为10 g·kg-1·d-1),模型组和正常对照组灌服相同剂量的生理盐水。通过HE染色、透射电镜观察再灌注6 h、12 h、24 h、48 h与72 h各组大鼠视网膜的形态学变化,原位杂交方法检测各组视网膜凋亡相关因子survivin的表达情况。结果与正常对照组相比,模型组再灌注后6 h视网膜神经节细胞层高度水肿,内丛状层明显增厚,部分视网膜神经节细胞发生空泡变性,但随时间延长病变逐渐减轻。治疗组各时间点视网膜损伤均较模型组轻。与正常对照组相比,模型组survivin在再灌注后6 h即开始增加,24 h达到高峰,以后逐渐下降,但各时间点均高于正常对照组(均为P<0.05)。与模型组相比,治疗组survivin各时间点表达均较高(均为P<0.05)。结论初步证实复方五花血藤对于大鼠视网膜缺血再灌注损伤有较为明显的保护作用,此研究对于临床药物的开发和利用具有积极意义。 Objective To explore the protective effects of compound stem of Sargentgloryvine on retinal ischemia reperfusion injury in rats. Methods Ninety healthy and clean SD rats were randomly divided into 3 groups: normal control group,model group and treatment group( lavaging the rats with a certain range of stem of sargentgloryvine,herb of tuberculate speranskia, spinyleaf pricklyash root and so on),and sub-divided into 6-hour group,12-hour group,24-hour group, 48-hour group and 72-hour group according to different reperfusion period. The morphologic change of each layer of cells in the retina was studied by HE and transmission electron microscope.The preoperative and postoperative positive expression of apoptotic related factor survivin was detected by the method in situ hybridization. Results After increasing intraocular pressure for 6 hours,the histology started to change,manifest as edema of gangliocyte and inner plexiform layer, few gangliocyte were vacuolar degeneration,but it recovered gradually with the extension of time,the normal control group showed normal retinal structure,while the treatment group gradually recovered from ischemic reperfusion,but was less than the same stage of ischemia-reperfusion group. After reperfusion for 12 hours,the positive expression of survivin started to increase,peaked at 24 hours, and then gradually decreased( P &lt; 0. 05). Compared with the model group,the positive expression of survivin in the treatment group decreased( P &lt; 0. 05). Conclusion The preliminary results show that compound stem of Sargentgloryvine drug has the obvious protective effects on retinal ischemia reperfusion injury,which has the positive significance on the development and utilization of clinical drug.
