HLA-G 与慢性炎症性疾病的研究进展 被引量:3

摘要 非经典人类白细胞抗原G( human leukocyte an-tigen-G, HLA-G)是机体内重要的免疫耐受分子,经选择性剪切可产生4种膜结合型 HLA-G 蛋白( mHLA-G,HLA-G1~G4)和3种可溶型HLA-G蛋白( sHLA-G, HLA-G5~G7)。相对于 HLAⅠ类分子,HLA-G具有有限的基因多态性,且存在多个位点和sHLA-G表达水平显著相关,其中研究最多的是14 bp插入/缺失等位基因多态性(14 bp+/-多态性)。当HLA-G基因为14 bp插入纯合子时,外显子8起始区的一个92 bp片段将被剪切,从而有助于HLA-G mRNA转录水平更稳定。但HLA-G转录水平与蛋白表达存在不均一性,中国汉族人种基因型为14 bp+/+人群外周血 sHLA-G 较14 bp+/-、14 bp-/-人群外周血sHLA-G分泌水平显著降低,进一步提示HLA-G 14 bp+/-多态性在sHLA-G分泌过程中发挥了至关重要的作用[1]。
