摘要 目的探讨rhIL-1α和rhTNF-α对人成纤维样滑膜细胞(FLS)PGE2受体(EP2)及Gαs表达的影响及芍药苷(Pae)的作用。方法体外培养人FLS,使用不同浓度的rhIL-1α和rhTNF-α刺激人FLS,观察EP2及Gαs蛋白表达的情况,观察Pae对其的影响。采用免疫荧光标记法结合激光共聚焦显微镜分析技术检测人FLS的EP2表达,Western blot检测Gαs表达。结果 rhIL-1α(0.1、1、10μg/L)和rhTNF-α(0.2、2、20μg/L)明显降低人FLS EP2和Gαs表达,Pae(10-8、10-7、10-6、10-5mol/L)可上调人FLS的EP2和Gαs表达。结论 Pae恢复rhIL-1α和rhTNF-α刺激后EP2和Gαs的表达可能是其抑制滑膜细胞增殖和亢进分泌功能的分子机制之一。 Objective To investigate the effects of paeoniflorin (Pae) on expression of EP2 and Gαts of human fibroblast-like synoviocytes (FLS) under the stimulation of rhIL-1 α and rhTNF-α.Methods FLS of human were cultured in vitro.The expression of EP2 on human FLS under the stimulation of rhIL-1 α and rhTNF-α and the effect of Pae were observed by immunofluorescence,analyzed by confocal laser microscopy,the expression of Cαs was detected by Western blot.Results rhIL-1α (0.1,1,10 μg/L) and rhTNF-α (0.2,2,20 μg/L) significantly inhibited the expression of EP2 and Gαs,Pae (10-8,10-7,10-6,10-5 moL/L)could improve the expression of EP2 and Gαs.Conclusion Improving the abnormal expression of EP2 and Gas on human FLS under the stimulation of rhIL-1 α and rhTNF-α is probablly one of the most important molecule mechanisms of Pae preventing RA.
